巴拉圭茶对高脂膳食小鼠氧化还原状态的影响

Effects of Ilex paraguariensis on Redox Status in High-fat Diet Fed Mice

  • 摘要: 目的:探究不同剂量巴拉圭茶对高脂膳食小鼠组织氧化还原状态的影响。方法:选取6周龄C57BL/6J雄性小鼠48只,随机分为四组,对照组(ND,基础日粮),高脂组(HF,高脂日粮),高脂+低剂量巴拉圭茶组(HFL,高脂日粮,按1 g/kg剂量灌胃巴拉圭茶),高脂+高剂量巴拉圭茶组(HFH,高脂日粮,按2 g/kg剂量灌胃巴拉圭茶)。6周后处死小鼠,测定小鼠肝脏氧化还原状态指标,肝脏FoxO3a、catalase、MnSOD基因表达水平。结果:与ND组相比,HF组小鼠肝脏活性氧自由基(Reactive oxygen species,ROS)水平、丙二醛(Malondialdehyde,MDA)含量显著增加(P < 0.05),过氧化氢酶(Catalase,CAT)活性、超氧化物歧化酶(Superoxide dismutase,SOD)活性、谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)活性、谷胱甘肽/氧化型谷胱甘肽(Glutathione/Oxidized glutathione,GSH/GSSG)比值、总抗氧化能力(Total antioxidant capacity,T-AOC)水平显著降低(P < 0.05),肝脏FoxO3a、catalase、MnSOD基因表达量显著下降(P < 0.05);与HF组相比,HFL 及HFH小鼠肝脏ROS水平、MDA含量显著降低(P < 0.05),CAT活性、SOD活性、GSH-Px活性、GSH/GSSG比值、T-AOC水平显著升高(P < 0.05),肝脏FoxO3a、catalase、MnSOD基因表达量显著提高(P < 0.05)。相关性分析表明,ROS与FoxO3a、catalase、MnSOD基因表达量成极显著负相关(P < 0.01),CAT、SOD、GSH-Px活性与FoxO3a、catalase、MnSOD基因表达量成极显著正相关(P < 0.01)。结论:适量的巴拉圭茶能改善高脂膳食造成小鼠氧化还原状态失衡的现象,这与FoxO3a基因通路有一定关系。

     

    Abstract: Objective: To investigate the effects of Ilex paraguariensis on redox status in high-fat diet fed mice. Methods: Forty-eight male C57BL/6J mice of six-week-old were randomly divided into 4 groups, normal diet (ND), high-fat diet (HF), high-fat diet+1 g/kg Ilex paraguariensis (HFL), high-fat diet+2 g/kg Ilex paraguariensis (HFH). The mice were sacrificed six weeks later. Hepatic redox status-related indicators were measured. The expression of FoxO3a, catalase and MnSOD genes in liver were analyzed. Results: Compared with the ND group, the levels of ROS and MDA in the liver of the HF group were increased significantly (P < 0.05), CAT, SOD, GSH-Px, GSH/GSSG values and T-AOC were reduced significantly (P < 0.05), the expression of FoxO3a, catalase and MnSOD genes in the liver were decreased significantly (P < 0.05). Compared with the HF group, the levels of ROS and MDA in the liver of the HFL and HFH groups were reduced significantly (P < 0.05), CAT, SOD, GSH-Px, GSH/GSSG values and T-AOC were increased significantly (P < 0.05), the expression of FoxO3a, catalase and MnSOD genes in the liver were increased significantly (P < 0.05). Correlation analysis showed that ROS and FoxO3a, catalase, MnSOD genes expression were significantly negative correlation (P < 0.01), CAT, SOD, GSH-Px and FoxO3a, catalase, MnSOD genes expression were significantly positive correlation (P < 0.01). Conclusion: Appropriate amount of Ilex paraguariensis can improve the redox state imbalance of mice caused by high-fat diet, which has a certain relationship with FoxO3a gene pathway.
