刺玫果中花色苷提取工艺优化及抗氧化性分析

Optimization of Extraction Technique and Antioxidant Activity of Anthocyanins from Rosa davurica Pall.

  • 摘要: 为了优化刺玫果中花色苷提取工艺并考察其抗氧化活性,本研究采用超声波辅助提取法从刺玫果中提取花色苷,通过单因素实验、Placket-Burman设计并结合Box-Behnken实验确定刺玫果中花色苷最佳提取条件;采用清除DPPH·的能力、超氧阴离子自由基(\rmO_2^-\cdot )能力、ABTS+·能力等多种体外抗氧化方法评价刺玫果中花色苷的抗氧化活性。结果表明:刺玫果中花色苷最佳提取条件为:料液比1:15 g·mL−1、超声温度50 ℃、乙醇体积分数60%、超声时间30 min,溶液pH为2,超声提取次数2次,刺玫果中花色苷含量为185.033 mg·100 g−1;体外抗氧化活性研究表明,刺玫果中花色苷对DPPH·清除能力IC50值为2.299 mg·mL−1、对超氧阴离子自由基清除能力IC50值为1.902 mg·mL−1、对ABTS+·清除能力IC50值为8.993 mg·mL−1。本实验确定的工艺简单易行,从刺玫果中提取的花色苷含量较高,体外多种抗氧化评价方法表明刺玫果中花色苷具有一定的抗氧化活性。

     

    Abstract: In order to optimize the extraction conditions of anthocyanins from Rosa davurica Pall. and investigate its antioxidant activity, single-factor experiment, Placket-Bumrman design, Box-Behnken analysis associated with ultrasonic extraction method were adopted. The antioxidant activity of anthocyanins from Rosa davurica Pall. were investigated by various methods, such as removal DPPH radical, \rmO_2^-\cdot radical, ABTS+ racial. The results showed that the optimum extraction conditions were as follows: Solid-liquid ratio was 1:15 g·mL−1, ultrasonic temperature was 50 ℃, ethanol volume was 60%, ultrasonic time was 30 min, pH was 2, and extraction times was 2. The content anthocyanins of Rosa davurica Pall. was 185.033 mg·100 g−1. The antioxidant activity was measured with IC50 and the results showed that the removal ability to DPPH free radical, superoxide anion free radical, ABTS+ free racial of the anthocyanins was 2.299, 1.902, 8.993 mg·mL−1, respectively. The technology in the current study was easy and feasible. The content of anthocyanins was considerable. Furthermore, it showed that the anthocyanins had certain antioxidant activity in various antioxidant experiments.
