欧李发酵液对高脂血症SD大鼠血脂的调控及对肝脏的保护作用

Effects of Cerasus humilis Fermentation Juice: Regulating Serum Lipids and Protecting Liver in Hyperlipidemia SD Rats

  • 摘要: 研究欧李发酵液(Cerasus humilis fermentation juice,CF)对高脂血症SD大鼠血脂的调控作用及肝保护作用。健康雄性SD大鼠30只,适应性喂养7 d后,依据体重随机分为对照组、高脂模型组与CF组(发酵液浓度为40%,灌胃剂量为10 mL/kg bw),每组10只,经口灌胃,1次/d,连续10周。灌胃期间于第0、2、4、6、8、10周末眼眶静脉丛采血,全自动生化分析仪测定血清总胆固醇(Total cholesterol,TC)、甘油三酯(Triglyceride,TG)、低密度脂蛋白胆固醇(Low density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(High density lipoprotein cholesterol,HDL-C)浓度;肝脏冰冻切片油红O染色观察肝脂滴情况并计算脂滴面积比;HE染色观察肝细胞病变情况。结果表明:大鼠的血脂随着造模时间的延长呈波动趋势。第8周时,与模型组相比,CF组血清TG浓度显著降低57.20%(P<0.05)。实验结束时,与模型组相比,CF组大鼠血清TC浓度降低了10.02%(P>0.05),血清LDL-C浓度显著降低35.48%(P<0.05)且与对照组浓度相近;CF组肝脏器指数较模型组显著降低了20.92%(P<0.05);肝油红脂滴面积比显示,CF组较模型组显著下降27.50%(P<0.05);HE染色结果显示CF组肝细胞脂质空泡明显减少。结果表明,欧李发酵液具有调控高脂血症SD大鼠血脂紊乱的作用,并能明显改善肝脏因脂质沉积而导致的病变。

     

    Abstract: In order to evaluate the antihyperlipidemic and hepatoprotective effects of Cerasus humilis fermentation juice,a hyperlipidemia model of SD rats was established with high fat feed. After 7 days of adaptive feeding,thirty healthy male rats were randomly divided into three groups(each group 10 rats):Control group,high lipid model group and Cerasus humilis fermentation juice(CF)group(the concentration of fermentation juice was 40%,the gavage dose was 10 mL/kg bw). SD Rats were given by oral gavage,once a day,for 10 weeks. Blood samples were collected from the orbital venous plexus at the end of week 0,week 2,week 4,week 6,week 8 and week 10. The levels of total cholesterol(TC),triglyceride(TG),low-density lipoprotein-cholesterin(LDL-C)and high-density lipoprotein-cholesterin(HDL-C)in serum were measured by automatic biochemical analyzer. Frozen section of livers was stained with oil red O,to assess the accumulation of hepatic lipid droplets and calculate their area ratio. HE staining was performed to observe the histopathological changes of liver. The results showed that the blood lipids of rats fluctuated with the extension of the modeling time. Compared with the model group,the concentrations of TG in serum was reduced significantly by 57.20%(P<0.05)at the end of week 8. At the end of the experiment,compared with the model group,the TC concentration in serum of the CF group was reduced by 10.02%(P>0.05),and the LDL-C concentration in serum,which was similar to the control group,was significantly reduced by 35.48%(P<0.05). Besides,the organ index of liver and area ratio of lipid droplets of the CF group were significantly reduced by 20.92%(P<0.05)and 27.50%(P<0.05),respectively. The results of HE staining presented that the hepatocyte lipid vacuoles were evidently decreased. The experimental results comprehensively indicated that CF didn’t only have the effect of regulating dyslipidemia in hyperlipidemia SD rats,but also could significantly improve liver lesions due to lipid deposition.
