鸡胚肽锌螯合物的制备工艺及免疫调节功能研究

Preparation of Chicken Embryo Peptide Zinc Chelate and Its Immunoregulatory Function

  • 摘要: 以鸡胚蛋为原料,通过分步酶解法制备鸡胚肽。以螯合率及螯合物得率为指标,利用单因素实验结合响应面法优化制备鸡胚肽锌螯合物,通过小鼠免疫器官胸腺和脾脏质量指数探究鸡胚肽锌螯合物的免疫调节功能。结果表明,先用碱性蛋白酶后用胰蛋白酶将鸡胚蛋进行酶解,在螯合温度70 ℃,鸡胚肽与锌的浓度比为2.3:1,pH为8,螯合时间2.2 h的条件下,鸡胚肽与锌元素的螯合率最高,为48.98%±0.35%,此时螯合物得率为38.04%±0.26%,得率较高且稳定。按照此方法制备的富锌鸡胚肽能使胸腺和脾脏的质量指数明显增大,说明通过双酶酶解制备的鸡胚小肽与锌元素的螯合物具有一定的免疫调节功能,并呈现一定的量效关系。鸡胚肽锌螯合物在食源性螯合肽免疫功能方面具有一定的开发利用价值。

     

    Abstract: Chicken embryo peptide was prepared from chicken embryo egg by step enzymolysis. Taking the chelation rate and the yield of chelate as the indexes, the single factor experiment and response surface method were used to optimize the preparation of chicken embryo peptide zinc chelate. The results showed that, under the condition of chelating temperature 70℃, concentration ratio of chicken embryo peptide to Zn+ 2.3:1, pH8 and chelating time 2.2 h, the chelating rate of chicken embryo peptide and zinc element was the highest (48.98%±0.35%), the chelating rate was 38.04%±0.26%, the yield was high and stable. It could increase the mass index of thymus and spleen obviously, which showed that the chelate of chicken embryo peptide and zinc had a certain immune regulation function and a certain dose effect relationship. Chicken embryo peptide zinc chelate had certain development and utilization value in the immune function of food derived chelate peptide.
