不同提取方法的毛建草精油抗氧化动力学研究

Study on the Antioxidative Kinetics of Essential Oil of Dracocephalum rupestre Hance by Different Extraction Methods

  • 摘要: 为比较不同毛建草精油提取方法对其抗氧化性的影响,本文分别采用溶剂浸提法、水蒸气蒸馏法提取毛建草精油,然后通过改变毛建草精油初始浓度、温度对其清除DPPH·的动力学进行研究;通过比较毛建草精油及芦丁的抗氧化性,探索毛建草蒸馏精油与芦丁间对DPPH·清除的协同作用。结果表明:溶剂浸提法提取的毛建草精油的抗氧化能力强于水蒸气蒸馏产品,在其体积分数为2.5%时,浸提精油的清除率达到81.56%,而蒸馏精油为57.78%;溶剂提取精油和水蒸气馏出精油与DPPH·反应的速率都随温度升高、初始浓度增大而增大,实验条件下,浸提精油与蒸馏精油清除DPPH·反应的活化能Ea分别为8.11、32.66 kJ·mol-1,反应均接近二级反应动力学方程;毛建草蒸馏精油与芦丁清除DPPH·的IC50值依次为0.034、0.006 mg/mL,而其1:1混合物为0.012 mg/mL,明显高于二者的拟合值0.009 mg/mL,毛建草精油中的抗氧化成分与芦丁不存在协同作用,反而存在拮抗作用。本研究关于毛建草精油清除DPPH·自由基的动力学及其与芦丁之间的相互作用,为更好地利用和开发不同的精油产品,提供了借鉴与思路。

     

    Abstract: To compare the effect of extraction methods of essential oil from Dracocephalum rupestre Hance on antioxidative ability,solvent extraction and steam distillation were used to obtain essential oils. Through changing initial concentration of essential oils or the experimental temperature,DPPH· scavenging kinetics of essential oils were studied. Synergistic effect between the distillate essential oil and rutin during DPPH· scavenging process was explored by comparing the antioxidative activity of distillate essential oil with that of rutin. The results showed that the antioxidative activity of the essential oil extracted from solvent was stronger than that of product from steam distillation. At the experimental maximum concentration,the scavening rate of essential oil from solvent extraction achieved to 81.56% and that of distillate essential oil was 57.78%. The reaction rate to DPPH· of ssential oil obtained by solvent extraction and steam distillation increased with the increasing of temperature and the initial concentration. Under experimental conditions,the Ea values of DPPH· scavenging reactions of essential oil from solution extraction and from distillation were 8.11,32.66 kJ·mol-1,respectively. Both the scavenging reactions were close to the second-order reaction kinetics equation. The IC50values of DPPH· scavenging reaction of distillation essential oil and rutin were 0.034 and 0.006 mg·mL-1,respectively,and the 1∶1 mixture was 0.012 mg·mL-1,which was significantly higher than the fitting values of the two. The antioxidative components in distillate essential oil of Dracocephalum rupestre Hance and rutin were not synergistic but mutually restricted. DPPH· scavenging kinetics of the essential oils and interaction between components in essential oil with rutin provide reference and ideas for better utilization and development of different essential oil products.
