山刺玫果多糖抗疲劳及抗氧自由基水平机制研究

Mechanism of Anti-fatigue and Anti-oxygen Free Radical Level of Polysaccharides from Rosa davurica Pall.

  • 摘要: 目的:研究不同剂量山刺玫果多糖对小鼠抗运动性疲劳作用及抗氧自由基水平的影响。方法:将150只雄性昆明小鼠分为3个实验组,每组小鼠随机分为空白对照组(给予等体积生理盐水)、人参水煎液阳性组(0.135 g·mL-1)、山刺玫果多糖高剂量组(100 mg·kg-1)、中剂量组(50 mg·kg-1)、低剂量组(25 mg·kg-1),按照0.1 mL·10 g-1剂量灌胃给药,通过测定小鼠力竭游泳时间、转棒时间,量化的比较了血乳酸(BLA)、肝糖原(LG)、肌糖原(MG)、血尿素氮(BUN)等的含量及肝脏与血清中总抗氧能力(T-AOC)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物歧化酶(GSH-Px)的活性、丙二醛(MDA)含量,综合评价了山刺玫果多糖的抗疲劳以及抗氧自由基的能力。结果:与空白组比较,山刺玫果多糖中剂量组和高剂量组的转棒时间显著延长(P<0.05);各剂量组的小鼠负重游泳时间均显著增加(P<0.05);中剂量组、高剂量组小鼠肌肉中MG及肝脏中LG含量均显著增加(P<0.05);高剂量组小鼠血清中BUN含量显著降低(P<0.05);中剂量组、高剂量组小鼠游泳后BLA浓度均显著降低(P<0.05);中剂量组及高剂量组小鼠肝脏中SOD活力显著提高(P<0.05),并且高剂量组小鼠血清中SOD活力显著提高(P<0.05);高剂量组小鼠肝脏中MDA含量显著降低(P<0.05),中剂量和高剂量组小鼠血清中MDA含量显著降低(P<0.05);中剂量、高剂量可使肝脏中T-AOC能力显著增加(P<0.05)。小鼠血清T-AOC活力测定结果中,低剂量组与中剂量组小鼠血清中T-AOC能力均显著增加(P<0.05);小鼠肝脏及血清GSH-Px活力测定结果中,高剂量组小鼠肝脏与血清中的GSH-Px的活力显著提高(P<0.05)。结论:山刺玫果多糖具有一定的抗疲劳和抗氧自由基作用。

     

    Abstract: Objective:To investigate the effects of polysaccharide on mice anti-fatigue effect of Rosa davurica Pall.fruit and anti-oxygen free radical level.Methods:A total of 150 Kunming male mice were randomly divided into three experimental group,and a blank control group(administered with equal volume of normalsaline)and a positive group of Ginseng decoction(0.135 g·mL-1),a high dose group of polysaccharides(100 mg·kg-1),medium dose group(50 mg·kg-1),low dose group(25 mg·kg-1),intragastric administration of 0.1 mL·10 g-1. Exhaustive swimming time,rotarod time,blood lactic acid(BLA),liver glycogen(LG),muscle glycogen(MG),blood urea nitrogen(BUN)contents and activity of total antioxidant capacity(T-AOC),superoxide dismutase(SOD)and glutathione superoxide dismutase(GSH-Px),malondialdehyde(MDA)contents in the liver and serum were measured,which were used to evaluate the anti-fatigue and anti-oxygen free radical ability of polysaccharide. Results:Compared with blank group,rotarod time were significantly prolonged in high and medium dose group(P<0.05).Weight-loaded swimming time of each dose group were significantly increased(P<0.05). The contents of MG and LG were significantly increased in middle-dose group and high-dose group. The contents of BUN in high-dose group were decreased. The content of BLA in high-dose group were significantly decreased(P<0.05)in middle-dose group and high-dose group(P<0.05).The activity of SOD in liver and serum of mice in high dose group were increased significantly(P<0.05)and the activity of SOD in liver of mice in medium dose group were increased significantly(P<0.05). The contents of MDA in serum of mice in medium dose group were decreased significantly,and that in liver and blood of mice in high dose group were decreased significantly(P<0.05). The ability of T-AOC were all increased in middle-dose group and high-dose group(P<0.05). The activity of GSH-Pxin liver and serum of high dose group were increased significantly(P<0.05). Conclusions:Polysaccharides from Rosa davurica Pall. would have good anti fatigue and anti oxygen free radical effects.
