锦橙中β-葡萄糖苷酶的提取纯化及其结构表征

Extraction,Purification and Structural Characterization of β-Glucosidase in Jincheng Oranges

  • 摘要: 本文以锦橙为原料,对其果皮与果肉中的β-葡萄糖苷酶进行了提取纯化以及结构表征。实验结果显示,果皮中0.35 mol/L NaCl洗脱液中的β-葡萄糖苷酶的平均酶活为(6.57±4.27)×10-3 U/g,其活性显著高于0.4 mol/L NaCl洗脱液中的β-葡萄糖苷酶活性和果肉中的0.35 mol/L和0.4 mol/L NaCl洗脱液中的酶活(p<0.05),且0.35 mol/L NaCl洗脱液中的蛋白质浓度最高,达0.154 mg/mL。在该条件下纯化得到的β-葡萄糖苷酶的分子量略高于170 kDa,且该酶的最大荧光吸收峰为582 nm,其粒径范围为5~25 nm。

     

    Abstract: In this paper,the β-glucosidase in peel and pulp of Jincheng oranges was extracted and purified. The results showed that the activity of β-glucosidase in 0.35 mol/L NaCl eluent from peel was (6.57±4.27)×10-3 U/g,which was significantly higher than that in 0.4 mol/L NaCl eluent,and it was also higher than that in 0.35 mol/L and 0.4 mol/L NaCl eluent from pulp (p<0.05). The protein content in 0.35 mol/L NaCl eluent from peel was also the highest at the level of 0.154 mg/mL. The molecular weight of this β-glucosidase was higher than 170 kDa,and the maximum fluorescence absorption was at 582 nm,and the size was about 5~25 nm.
