石榴籽多糖硒酸酯合成工艺优化及其抗油脂氧化能力

Optimization of Synthesis Technology of Selenium Polysaccharide Ester from Pomegranate Seeds and Its Anti-lipid Oxidation Ability

  • 摘要: 本文以石榴籽多糖和亚硒酸钠为原料,以合成产物硒含量和收率为考察指标,原料配比、反应温度、硝酸浓度和反应时间为试验因子,通过单因素实验和Box-Behnken试验设计方法对石榴籽多糖硒酸酯合成工艺条件进行优化,并对合成产物的结构和抗油脂氧化能力进行分析。结果显示,石榴籽多糖硒酸酯的最佳合成工艺条件为:原料配比1:1.02,反应温度76 ℃,硝酸浓度0.74%,反应时间9.3 h,在此工艺条件下,合成产物硒含量为(4.117±0.02) mg/g,产物收率为42.596%±0.13%,紫外光谱、红外光谱和热重分析充分证实石榴籽多糖硒酸酯中含有Se=O键、Se-O键和Se-C键;合成产物具有一定的抗油脂氧化能力,能够明显减缓油脂POV值升高趋势,且有一定的浓度依赖性。该研究为石榴籽资源的充分开发利用提供理论依据。

     

    Abstract: Polysaccharide of pomegranate seeds and sodium sulfite as raw materials,contents of Se in synthetic product and yields of synthetic product as investigation index,raw material ratio,reaction temperature,concentration of HNO3 and reaction time as experimental factors,the synthesis conditions were optimized by single factor test and Box-Behnken experimental designs,and simultaneously the product structure and antioxidant capacity were studied. The results showed that,the optimum preparation conditions of selenium polysaccharide ester of pomegranate seeds were as follows:Raw material ratio was 1:1.02,reaction temperature was 76 ℃,concentration of HNO3 was 0.74%,and reaction time was 9.3 h. Under this conditions,contents of Se in synthetic product was(4.117±0.02) mg/g,and yields of synthetic product was 42.596%±0.13%. UV,IR and TGA analysis fully confirmed that selenium polysaccharide ester of pomegranate seeds contained Se=O,Se-O and Se-C. Synthetic product had a certain antioxidant capacity,so it could significantly slow the trend of oil POV value increasing trend,and had a certain amount of concentration-dependent manner. The study provided a theoretical basis for the full development and utilization of pomegranate seeds resources.
