霍山石斛与铁皮石斛的体外生物活性比较

Comparison of Biological Activities in Vitro of Dendrobium huoshanense and Dendrobium officinale

  • 摘要: 目的:比较霍山石斛与铁皮石斛(乐清、云南)3种石斛提取物对体外免疫、抗氧化、细胞氧化损伤、抗肿瘤和抑制α-葡萄糖苷酶活性的影响。方法:将3种石斛水提物配置成不同浓度溶液,分别与对照品一起进行体外测定刺激巨噬细胞RAW264.7释放NO的活性,清除ABTS自由基、FRAP法抗氧化能力和阻止H2O2诱导PC12细胞衰老能力,抑制肝癌细胞HepG2活性和胰岛素抵抗HepG2模型细胞内α-葡萄糖苷酶的活性研究。结果:霍山石斛提取物具有最优的体外刺激RAW264.7细胞释放NO的活性,更好的ABTS自由基清除能力和FRAP能力。200 μg·mL-1霍山石斛提取物对H2O2诱导的PC12细胞具有保护作用,修复率为40.52%。此外3种石斛提取物均具有一定的抑制HepG2肿瘤细胞增值的活性。与胰岛素抵抗模型组相比,在低浓度(100 μg·mL-1)时,3个样品均对细胞内α-葡萄糖苷酶的活性无抑制作用,在中高浓度时霍山石斛提取物(200和400 μg·mL-1)具有一定的抑制活性,表现出较好的降血糖作用。结论:霍山石斛相较于铁皮石斛在增强免疫、抗肿瘤、抗氧化、延缓衰老及降血糖等体外生物活性方面具有更好的作用。

     

    Abstract: Objective:This study was designed to compare biological activities of Dendrobium. huoshanense and Dendrobium officinale(Yueqing,Yunnan)in vitro. The immunity activities,antioxidant activities,cell oxidative stress injury,antitumor activities and α-glucosidase inhibitoryassay were analyzed. Method:Three kinds of Dendrobium water extracts with different concentration were used to activity experiments in vitro,including the release of NO by stimulating macrophage RAW264.7,the ability of clearing ABTS free radicals,the total antioxidant capacity with FRAP method,the ability of preventing the cell PC12 aging induced by H2O2,the activity of hepatoma cell HepG2 and the activity of intracellular α-glucosidase of insulin resistance HepG2 model. Results:The results showed that the extract of Dendrobium huoshanense could significantly stimulate macrophages to release NO in vitro and strongest antioxidant of ABTS scavenging ability and Ferric ion reducing power. The Alamar Blue results indicated Dendrobium huoshanense extracts at the concentration of 200 μg/mL had better protective effect on activity decrease of PC12 cell induced by H2O2,and the recovery ratio was 40.52%. In addition,the water extracts from 3 species of Dendrobium could inhibit in growth of HepG2 cells. The Dendrobium huoshanense extracts(200,400 μg·mL-1)had stronger inhibitory effect on α-glucosidase than other fragments in insulin resistant model HepG2 cells,while water extracts from 3 species of Dendrobium had no activity at 100 μg·mL-1. Therefore,Dendrobium huoshanense had considerable anti-hyperglycemic activities. Conclusion:Dendrobium huoshanense had a better effect than Dendrobium officinale in enhancing biological activity such as immunity,anti-tumor,anti-oxidation,anti-aging and blood sugar lowering.
