Abstract:
The insulin resistant models of HepG2 cells were induced by insulin to study the effects of D-pinitol compound with Mn
2+ on glucose consumption and glycogen synthesis. The expression of AMPK genes was detected with real-time polymerase chain reaction(RT-PCR). The results showed that when the insulin concentration was 1×10
-3 mmol/L and the action time was 36 h,the glucose consumption reached the minimum. Compared with the model group,when the D-pinitol concentration was 100 mg/L and MnSO
4 was 10 mg/L,glucose consumption and glycogen content were extremely significantly(
p<0.01). The mRNA expression levels of
AMPKα-1,
AMPKα-2 and
GLUT4 increased significantly(
p<0.05)while
G6pase mRNA expression decreased significantly(
p<0.05). D-pinitol compound with Mn
2+ could improve the glucose consumption,increase glycogen synthesis. Moreover,it had a positive effect on hypoglycemic activity through AMPK signaling pathway.
