两种魔芋多糖抗疲劳及降血糖活性

Anti-fatigue and Anti-hyperglycemic Activities of Two Konjac Polysaccharides

  • 摘要: 目的:研究不同剂量的魔芋葡甘露聚糖(Konjac glucomannan,KGM)和魔芋葡甘露低聚糖(Konjac glucomannan oligosaccharide,KOGM)对小鼠抗疲劳作用及降血糖活性的影响。方法:将35只无特定病原体(Specific pathogen free,SPF)级昆明种雄性小鼠随机分为空白对照组(给予等体积生理盐水)、魔芋葡甘露聚糖高剂量组(60 mg/kg)、中剂量组(30 mg/kg)、低剂量组(15 mg/kg)以及魔芋葡甘露低聚糖高(60 mg/kg)、中(30 mg/kg)、低(15 mg/kg)剂量组共7组,每组5只。通过负重游泳实验来记录小鼠的力竭游泳时间,并通过测定超氧化物歧化酶(SOD)、丙二醛(MDA)等相关生化指标,测定魔芋葡甘露聚糖及其低聚糖的抗疲劳作用。另将40只小鼠随机分为空白对照组(等体积生理盐水)、阳性对照组(盐酸二甲双胍110.5 mg/kg)和魔芋葡甘露聚糖及其低聚糖高、中、低剂量组,共8组,每组5只。通过口服葡萄糖糖耐量实验(Oral glucose tolerance test,OGTT),并分别测定小鼠口服葡萄糖0、30、60、120 min后的血糖值,观察魔芋葡甘露聚糖及其低聚糖对正常小鼠糖耐量的改善作用。结果:在抗疲劳实验中,与空白组相比KGM和KOGM各剂量可以在一定程度上延长小鼠负重游泳时间,同时增加小鼠肝脏中的SOD活力,降低MDA含量,且以高剂量组最为显著(p<0.05);在糖耐量实验中,两种不同分子量的多糖均具有降糖效果,且以高剂量组效果最佳。结论:魔芋葡甘露聚糖及其低聚糖具有良好的抗疲劳作用,同时具有良好的降血糖,改善糖耐量作用。

     

    Abstract: Objective:To study the effects of different doses of konjac glucomannan(KGM)and konjac glucomannan oligosaccharide(KOGM)on anti-fatigue and hypoglycemic activity in mice. Methods:A total of 35 Kunming male mice with no specific pathogen free(SPF)grade were randomly divided into a blank control group(administered with equal volume of normal saline)and a high dose group of konjac glucomannan(60 mg/kg),medium dose group(30 mg/kg),low dose group(15 mg/kg),and konjac glucomannan oligosaccharide high(60 mg/kg),medium(30 mg/kg),low(15 mg/kg)group. There were 7 groups in the dose group,5 mice in each group. The weight swimming test was used to record the exhaustive swimming time of mice,and the anti-oxidation activity of konjac glucomannan and its oligosaccharides was determined by measuring the biochemical indicators such as superoxide dismutase(SOD)and malondialdehyde(MDA). Another 40 mice were randomly divided into a blank control group(equal volume of normal saline),a positive control group(metformin hydrochloride 110.5 mg/kg)and konjac glucomannan and its oligosaccharide high,medium and low dose groups,8 groups of 5 mice each. Oral glucose tolerance test(OGTT)was used to measure the blood glucose levels of mice after oral glucose for 0,30,60,120 min,and effect of konjac glucomannan and its oligosaccharides on improving glucose tolerance were observed in normal mice. Results:In the anti-fatigue experiment,compared with the blank group,the doses of KGM and KOGM could prolong the weight-bearing swimming time of mice to some extent,increase the SOD activity in the liver of mice,reduce the MDA content,and the highest dose group was significant(p<0.05);in the glucose tolerance experiment,two different molecular weight polysaccharides had hypoglycemic effect,and the best effect was in the high dose group. Conclusion:Konjac glucomannan and its oligosaccharides had good anti-fatigue effects,and had good blood sugar lowering effect and improved glucose tolerance.
