黑曲霉诱导子促进青钱柳细胞合成三萜的氧化应激机制

Incentive Mechanism of Triterpenoid Synthesis by Antioxidant Response in the Cultured Cyclocarya paliurus Cells under the Elicitation of Aspergillus niger Elicitor

  • 摘要: 本文旨在研究黑曲霉诱导子(Aspergillus niger elicitor,ANE)促进青钱柳细胞合成三萜类化合物的氧化应激机制。通过研究抗氧化应激相关酶活性的变化和相关基因的表达量,从氧化应激层面诠释ANE的诱导作用机制。结果表明,在ANE诱导后10 h时,青钱柳细胞的三萜合成量开始升高,至60 h时达到最高值,三萜含量达36.46 mg/g,为对照组的3.36倍。在ANE刺激之下,超氧化物歧化酶(Superoxide Dismutase,SOD)、过氧化物酶(Peroxidase,POD)、过氧化氢酶(Catalase,CAT)、苯丙氨酸解氨酶(phenylalanin ammo-nialyase,PAL)、磷脂酶C(Phospholipase C,PLC)五类酶的活性均显著升高(p<0.05),且先后出现了数个酶活力峰值。转录组测序结果表明,3个SOD、20个POD、1个CAT、3个PAL、3个PLC相关基因的表达量升高,其中3个POD、1个CAT、3个PAL相关基因的表达量显著上升,此外,细胞中三萜合成酶相关基因表达亦发生上调。由此可以推断,在ANE刺激下青钱柳细胞出现氧化应激,导致抗氧化酶相关基因表达量的升高,相应酶的活性因此上升,进而引起细胞发生防御性反应,最终促进了细胞内三萜的合成。本文从氧化应激层面探索ANE促进细胞合成三萜的作用机制,为植物细胞培养生产活性次级代谢产物提供了理论依据。

     

    Abstract: This paper was aimed to investigate the incentive mechanism of triterpenoid synthesis by antioxidant response in the cultured Cyclocarya paliurus cells under the elicitation of Aspergillus niger elicitor(ANE),and the activity changes of the relative antioxidant enzymes and the expression of related genes were investigated to explain the induction mechanism of ANE from oxidative stress level.Results showed that,the triterpenoid synthesis began to rise in the 10th h after inoculation and reached the peak in the 60th hour. The maximum triterpenoid content was 36.46 mg/g,which was 3.36 times as much as that of control. Meanwhile,the activity of SOD,POD,CAT,PAL,and PLC increased significantly,and several peaks of enzyme activity emerged successively in the cells under ANE stimulation. Moreover,the results of transcriptome detection showed that there were a number of up-regulated related genes including 3 SOD,20 POD,1 CAT,3 PAL,and 3 PLC,among which 3 POD,1 CAT,and 3 PAL were significantly up-regulated(p<0.05). Besides,the transcript abundance of related genes encoding triterpenoid synthetase increased as well. Consequently,it could be deduced that antioxidant response occurred in C. paliurus cells under ANE elicitation,which resulted in the increasing of expression and enzyme activity of genes related to oxidative stress and further improved the triterpenoid synthesis in the cells. The current paper investigated the incentive mechanism of triterpenoid synthesis by antioxidant response in the cultured Cyclocarya paliurus cells under the ANE elicitation,which provided reference for production of secondary metabolites by plant cell culture.
