葡萄状枝瑚菌菌丝液体培养条件优化

Optimization of Liquid Culture Conditions of Mycelium of Ramaria botrytis

  • 摘要: 为了研究液体培养条件对葡萄状枝瑚菌菌丝生长量的影响,通过单因素实验和正交试验分别考察了碳源、氮源、无机离子组合、生长因子、培养基初始pH、培养温度、摇床转速和光照这8个因素对菌丝生长量的影响,结果表明,最佳培养条件为:在23℃、摇床转速为220 r/min时,培养基初始pH6.5,装瓶量50 mL/250 mL锥形瓶,接种量10%(V/V),全黑暗培养7 d,1 L液体培养最佳培养基为25 g可溶性淀粉,3.5 g酵母粉,4.4 g(1.2 g KH2PO4+1.6 g FeSO4·7H2O+1.6 g MgSO4·7H2O)无机离子,0.01 g VB1,在此条件下葡萄状枝瑚菌菌丝生长量为(26.8±0.29)g/L。此研究结果为葡萄状枝瑚菌的液体发酵奠定了基础。

     

    Abstract: In order to study the effect of liquid culture conditions on mycelial growth of Ramaria botrytis,eight factors included carbon source,nitrogen source,inorganic ion combination,growth factor,initial pH of culture medium,culture temperature,shaking speed and light were investigated by single factor and orthogonal experiment. The results showed that,the optimum culture conditions were as follows:At 23℃ and shaking speed of 220 r/min,initial pH6.5,flask volume 50 mL/250 mL erlenmeyer flask,inoculum density 10%(V/V),full darkness culture for 7 days,the optimum liquid culture medium for 1L was 25 g soluble starch,3.5 g yeast extract,4.4 g(1.2 g KH2PO4+1.6 g FeSO4·7H2O+1.6 g MgSO4·7H2O)inorganic ion combination and 0.01 g VB1,under the above conditionsthe mycelial growth of R. botrytis was(26.8±0.29) g/L,the result laid the foundation for liquid fermentation of R. botrytis.
