文冠果种皮黑色素的提取及其结构表征

Extraction and Structural Characterization of Melanin from Testae of Xanthoceras sorbifolia

  • 摘要: 以NaOH溶液为提取溶剂,盐酸水解结合多种有机溶剂洗涤的方法,从文冠果种皮中提取纯化植物黑色素。研究了NaOH浓度、提取温度、提取时间以及液料比对提取效果的影响,并采用化学定性分析、紫外光谱、红外光谱和电子顺磁共振,对纯化文冠果种皮黑色素的结构及理化性质进行了研究。结果表明,适宜的提取工艺为:1 mol/L NaOH在50 ℃下提取2次,每次12 h,液料比15:1 mL/g。在此条件下,黑色素粗提物得率为14.42%,进一步经过酸水解和有机溶剂洗涤,纯化黑色素得率为5.23%,纯化后400 nm处色价提高了21.0%。文冠果种皮黑色素在理化性质方面与合成多巴黑色素非常相似:不溶于水和一般有机溶剂,在1 mol/L KOH溶液中完全溶解,pH<3时发生沉淀,可被NaClO和H2O2氧化漂白,多酚定性反应呈阳性;在紫外-可见光区没有吸收峰,红外光谱中在3400 cm-1附近和1650~1620 cm-1各有一个强的吸收带,顺磁共振波谱为典型的单线一次微商波谱,无超精细结构。

     

    Abstract: Melanin from testae of Xanthoceras sorbifolia was extracted and purified by alkaline extraction, acid hydrolysis and organic solvent wash. The effects of solvent concentration, temperature, time, and liquid/solid ratio on the extraction were studied. The melanin was characterized by diagnostic tests and spectroscopies of ultraviolet visible light (UV-Vis), infrared spectrum (IR) and electron paramagnetic resonance (EPR). Results showed that the melanin was suitable to be extracted with 1 mol/L NaOH and liquid/solid ratio of 15 mL/g at 50℃ for 12 h, twice. Under these conditions, the yield of melanin crude extract was 14.42%, and the yield of the purified melanin was 5.23% through further acid hydrolysis and organic solvent washing. After the purification, the color value at 400 nm was enhanced by 21.0%. The physicochemical properties of the melanin showed a strong similarity to these of the synthetic DOPA melanin. They were insoluble in both water and common organic solvents and dissolved in 1 mol/L KOH, precipitated below pH3, bleached in NaClO and H2O2, and the qualitative reaction of polyphenols were positive. The melanin absorption spectrum represented the decreasing line in the ultraviolet and visible range of wavelengths. The infrared spectrum had a broad absorption peak around 3400 cm-1 and a strong one at 1650~1620 cm-1. The EPR spectrum was a single line devoid of hyperfine splitting.
