高效抑制大肠埃希氏菌的芽孢杆菌的发酵培养基及发酵条件优化

Optimization of Fermentation Culture Medium and Conditions of Bacillus with High Antibacterial Activity against Escherichia coli

  • 摘要: 本研究从土壤中筛选对大肠埃希氏菌具有高抑菌活性的芽孢杆菌,采用单因素实验与正交试验,对发酵培养基及发酵条件进行优化。结果表明,经筛选得到一株对大肠埃希氏菌具有显著抑菌活性的芽孢杆菌G3E,经菌种鉴定为解淀粉芽孢杆菌植物亚种(Bacillus amyloliquefaciens subsp.plantarum);G3E表达抑菌物质的最佳发酵培养基配方为:淀粉10 g/L、氯化钙4 g/L、酵母浸出物4 g/L、胰蛋白胨4 g/L、尿素2 g/L;最佳发酵条件为:发酵温度40 ℃,发酵时间40 h,pH=7.0,此时,抑菌圈直径可达(25.5±0.5) mm。

     

    Abstract: Bacillus with inhibiting Escherichia coli function was screened from the soil. The fermentation medium and fermentation conditions were optimized by single factor and orthogonal test.The results showed that Bacillus G3E, which had strong bacteriostatic effect against Escherichia coli was screened and identified as Bacillus amyloliquefaciens subsp.plantarum. The best fermentation medium for G3E was starch 10 g/L, calcium chloride 4 g/L, yeast extract 4 g/L, tryptone 4 g/L, urea 2 g/L.The optimum fermentation conditions were as follows:The fermentation temperature was 40℃, the fermentation time was 40 h, the pH value was 7.0.At this time, the diameter of the bacteriostasis circle was (25.5±0.5) mm.
