蝙蝠蛾拟青霉Cs-4提取物对链脲佐菌素诱导2型糖尿病小鼠的降血糖作用及机制研究

Antidiabetic Effect and Its Underlying Mechanisms of Paccilomyces hepiali Cs-4 Aqueous Extract in Streptozotocin Induced Type Ⅱ Diabetic Mice

  • 摘要: 研究蝙蝠蛾拟青霉Cs-4提取物(PHAE)的降血糖活性及其作用机制。建立链脲佐菌素(STZ)诱导2型糖尿病C57BL/6J小鼠模型,考察PHAE灌胃处理对糖尿病模型小鼠糖脂代谢相关指标的影响。在此基础上,利用小鼠胰岛杂交瘤细胞MIN6初步探讨其作用机制。结果表明,与模型组相比,连续20 d灌胃PHAE(1000、3000 mg/kg)可显著提高糖尿病小鼠体重和血清胰岛素水平(p<0.05),并显著降低其空腹血糖、血清胆固醇、甘油三酯和低密度脂蛋白水平(p<0.05)。此外,PHAE可刺激MIN6细胞增殖、促进分泌胰岛素并且显著上调Epac2蛋白表达。实验结果提示,蝙蝠蛾拟青霉Cs-4提取物具有降血糖和调血脂活性,其降糖活性可能与其作用于胰岛细胞并促进胰岛素分泌有关,这一研究结果为蝙蝠蛾拟青霉的深入研究开发提供一定的数据依据。

     

    Abstract: This work aimed to investigate the hypoglycemic effect of Paccilomyces hepiali extract(PHAE)and its mechanism. Type 2 diabetic C57BL/6J mice model was induced by streptozotocin(STZ). The effects of PHAE on blood glucose and lipid level of diabetic mice were investigated. In addition,the hypoglycemic mechanism was also investigated in mice islets hybridoma cell line MIN6. Compared with the diabetic mice,PHAE(1000 and 3000 mg/kg)administration for 20 days led to a significant increase in bodyweight and serum insulin(p<0.05),reduction in fasting glucose levels,and the densities of serum cholesterol,triglyceride and low-density lipoprotein cholestrol(p<0.05). The results in vitro showed that PHAE stimulated cell proliferation and promoted MIN6 cells to release insulin and it also significantly increased Epac2 protein expression compared with untreated cells. Taken together,the results of the present work proved the hypoglycemic and hypolipidemic activity of Paccilomyces hepiali Cs-4 extract. The hypoglycemic effect of PHAE might be explained by promoting insulin secretion in pancreatic islet cells. The results of this study could provide useful data for furture development and application of Paeccilomyces hepiali.
