Molecularly imprinted polymer as artificial antibody in enzyme-linked immunosorbent assay for the determination of enrofloxacin

摘要: 在聚苯乙烯酶标板表面直接合成对恩诺沙星具有特异性识别吸附能力的分子印迹聚合物膜,将其作为仿生抗体代替生物抗体,建立直接竞争仿生酶联免疫分析法(ELISA)进行恩诺沙星的检测。结果表明最适条件下仿生酶联免疫方法的最低检出限(IC15)为 0.80 μg/L,灵敏度(IC50)为 65.93 μg/L,对恩诺沙星的代谢物环丙沙星有较高的交叉反应率(72.85%),对其它7种喹诺酮类结构类似物的交叉反应率均较低(0%~2.87%)。该方法对鸡肉组织和猪尿两种样品的添加回收实验中(鸡肉加标量分别为2.5、15、75 μg/kg;猪尿加标量分别为5、30、150 μg/kg),有较高的回收率(鸡肉:97.30%~103.56%;猪尿:94.48%~96.53%),采用HPLC方法与仿生ELISA方法进行对比验证无显著性差异(p>0.05)。该仿生ELISA法准确度、精密度及特异性均较高,步骤简单,与传统ELISA方法相比省时经济,可以应用于实际样品中恩诺沙星残留的快速检测。Abstract: The imprinted film specific to enrofloxacin was synthesized using polystyrene microtiter plate as support combining surface imprinting technique. A direct competition ELISA was developed with imprinted film as artificial antibody. The results showed that, the detection limit (IC15) and sensitivity (IC50) of the method were 0.80 and 65.93 μg/L, respectively. The specificity of the method was also investigated by comparing the cross-reactivity of other structurally related compounds with that of the template. The developed ELISA showed a low degree of cross-reactivity of these compounds (0%~22.87%) except ciprofloxacin (72.85%), which was the metabolite of enrofloxacin. The established ELISA has been applied in spiked chicken meat and swine urine samples with the recoveries ranging from 97.30% to 103.56% (the adding standard matter amount in chicken meat were 2.5, 15, 75 μg/kg) and 94.48% to 96.53% (the adding standard matter amount in swine urine were 5, 30, 150 μg/kg). The obtained results correlated well to those gained from HPLC method, which demonstrated that the developed ELISA method was reliable for rapid enrofloxacin detection in real samples.
