母乳挥发性成分固相微萃取-气质联用检测条件的优化及测定

Optimization of analysis conditions of volatile compounds in human breast milk by SPME-GC-MS

  • 摘要: 采用固相微萃取-气质联用分析母乳挥发性成分,并对其检测条件进行优化。通过单因素实验,最终确定了母乳挥发性成分较优的检测条件为使用DVB/PDMS萃取头、加入1.5 g NaCl的5 mL样品在60 ℃萃取30 min、解析5 min。在此检测条件下分析母乳样品,得到34种挥发性成分,其中烯烃类14种、酸类5种、酯类8种、芳香烃类4种、酮类3种,将为改善配方奶的风味提供一定的理论依据。

     

    Abstract: The combined technology of solid phase microextraction(SPME)and gas chromatography-mass spectrometry(GC-MS)was used to analyze the volatile compounds from human breast milk and the detection conditions were optimized by single factor experiments. The optimal conditions were achieved when 5 mL of sample containing 1.5 gram of NaCl was extracted at 60 ℃ for 30 min using DVB/PDMS extraction fiber with a resolution time of 5 min. Under optimal conditions,thirty-four volatile compounds including alkenes(fourteen types),acids(five types),esters(eight types),aromatic hydrocarbons(four types)and ketones(three types)were identified. This study could provide theoretical basis for the improvement of flavor of formula milk.
