响应面法优化黄芪下脚料蛋白提取工艺

Optimization of protein extraction from Astragali radix waste by response surface methodology

  • 摘要: 为确定黄芪下脚料蛋白的最佳提取工艺,有利于其进一步的活性研究,为其资源开发利用提供科学依据,使其变废为宝。通过单因素实验考察了提取温度、料液比、p H、提取时间和提取次数对黄芪下脚料蛋白提取率的影响,并通过响应面法优化了提取条件。结果表明,凯氏定氮法测得黄芪下脚料中蛋白含量为10.74%±0.13%,蛋白最佳提取工艺为提取温度64℃、液料比47∶1(m L·g-1)、p H12.0、提取时间4.3 h,提取2次,在此条件下,提取率的理论最大值为46.96%,实验验证值为47.03%±0.78%,采用响应面法优化得到黄芪下脚料蛋白提取工艺的参数准确可靠,该回归模型具有较好的预测性能,可用于指导生产实践,为黄芪下脚料蛋白的工业化生产提供一定的技术参考。 

     

    Abstract: In order to further research on its activity and providing scientific basis for its resource development and utilization, we determine the best extraction technology of Astragali radix waste protein, making waste into treasure of Astragali radix waste.The effects of extraction temperature, solvent/solid ratio, p H, extraction time and extraction times on the extraction rate of Astragali radix waste were investigated by single factor test.In addition, the extraction conditions were optimized by response surface methodology.Results showed that the protein content of Astragali radix waste was 10.74%±0.13%by kjeldahl method.The optimal extraction conditions of Astragali radix waste protein were as follows:the extraction temperature was 64℃, the solvent/solid ratio was 47∶1 (m L·g-1) , p H was 12.0 and extraction time was 4.3 h.Under this condition, the theoretical maximum of extraction rate was 46.96%, and the experimental value was 47.03%±0.78%.The extraction condition of Astragali radix waste protein optimized by response surface methodology was accurate and reliable and the regression model has good predictive performance, which can guide the production practice and provide some technical reference for industrial production of Astragali radix waste protein.
