大孔吸附树脂法分离纯化红景天中洛塞维

The isolation and purification of Rosavin by the macroporous adsorption resin

  • 摘要: 本文采用大孔吸附树脂法对红景天中洛塞维(Rosavin)分离纯化进行了研究,建立了如下工艺条件:Rosavin检测的高效液相(High performance liquid chromatography,HPLC)最佳的流动相为乙腈-0.2%磷酸(65/43,v/v),最佳柱温为30℃;选用非极性树脂D312,最大吸附量上样浓度为0.24 mg/m L,装柱高度为30 cm,粗品最大上样量为4 BV,上样流速为12 BV/h,洗脱剂为50%乙醇溶液。大孔吸附树脂分离纯化目的物Rosavin的含量由3.00%提高到了68.76%。大孔吸附树脂法分离纯化洛塞维的工艺条件合理,节约成本、收率高,适合工业应用。 

     

    Abstract: To establish a method for the Rosavin of isolation and purification, the result of process conditions were as follows:the conditions of HPLC test is acetonitrile-0.2% phosphoric acid ( 65/43, v/v) of mobile phase, and column temperature condition 30 ℃; on the comparison of six resin's adsorption and the establishment of the resin were preliminary isothermal static adsorption curve, we can determined the optimal resin was D312 resin, and the largest loading concentration was0.24 mg/m L; column height 30 cm, sample size 4 BV, flow rate 1 ~ 2 BV/h. The results showed that the Rosavin content had increased from 3.00% to 68.76%, based on the consideration of four factors in column chromatography process. The macroporous adsorption resin of isolation and purification of Rosavin is reasonable, it is suitable for industrial applications.
