萃取-吸附法分离萃取发酵液中红曲黄色素

Extraction and adsorption of Monascus yellow pigments from extractive fermentation broth

  • 摘要: 萃取发酵是红曲色素发酵生产的一种新技术,可促进胞内色素分泌到胞外,但存在发酵液中红曲色素与萃取剂(表面活性剂Triton X-100)难以分离纯化的问题。本研究采用三氯甲烷萃取结合碱性S-8树脂吸附的方法,分离了发酵液中黄色素和表面活性剂。通过该方法色素的回收率达到64.5%,表面活性剂的去除率为97.47%。在有机溶剂中,树脂对黄色素吸附的过程符合Freundlich热力学吸附平衡模型和准二级动力学模型。吸附的黄色素用含0.5%(w/v)盐酸的甲醇溶液被有效洗脱,总洗脱率达到79.48%。该方法为萃取发酵液中红曲黄色素的有效分离纯化提供了新途径。 

     

    Abstract: Extractive fermentation was an advanced technology for a high-yield productivity of Monascus pigments.Intracellular pigments could be extracted to the extracellular broth with surfactant Triton X-100. However, the solid bonding between pigments and Triton X-100 micelles challenged a subsequent separation and purification of the pigments from fermentation broth.This study demonstrated a selective isolation of Monascus yellow pigments from the extractive fermentation broth through a dual-phase chloroform extraction and subsequent adsorption using S-8 alkali resin.Recovery of the pigments and efficiency of Triton X-100 removal reached 64.5% and 97.47%, respectively. The pigment adsorption could be described by Freundlich equilibrium model with a pseudo-second order kinetic course.The use of methanol solution containing 0.5% ( w/v) HCl led to the highest elution efficiency with a total pigment recovery yield of 79.48%.The method was demonstrated to be a new approach for pigment isolation in extractive fermentation.
