蛹虫草基质多糖纳米硒复合物的制备研究

Study on the preparation of Cordyceps militaris polysaccharide/Nano-Selenium complex

  • 摘要: 以多糖为软模板,可以制备高效低毒的纳米硒-多糖复合物。本文以废弃的蛹虫草培养基质中提取的水溶性多糖为原料,研究了蛹虫草基质多糖纳米硒复合物的制备工艺及抗肿瘤活性。实验结果表明,在多糖浓度为10 mg/m L、亚硒酸浓度为0.4 mol/L、混合时间为4 h、滴加速度为1.5 r/min的条件下,可以制备出形貌理想、粒径适宜、稳定性良好的蛹虫草基质多糖纳米硒复合物。该复合物对Caco-2肿瘤细胞的生长具有一定抑制效果,且对细胞毒作用呈现出剂量依赖效应。 

     

    Abstract: Polysaccharides was used as soft template to prepare polysaccharide/Nano-Selenium complexes, which had high biological activity and low toxicity.The preparation procedure of Nano-Se/Cordyceps militaris stroma polysaccharides complexes was investigated by single-factor experiment and orthogonal experimental design, and the water-soluble polysaccharide which was exacted from the abandoned cordyceps militaris culture medium was used as the raw material. The results showed that the optimal reaction conditionswere as follows: the polysaccharide concentration was 10 mg/m L, selenite concentration was 0.4 mol/L, the mixing time was 4 hours, the preparation conditions of dropping was 1.5 r/min. Under the optimal condiction, the ideal morphology, ideal particle size, stable and good nano selenium/Cordyceps militaris stroma polysaccharide complex particles ( CMSP-Se NPs) could be successfully prepared.The antitumor experiment results showed that CMSP-Se NPs could inhibit the proliferation of Caco-2 cells, which presented a certain dose-response relationship.
