不同提取方法猴头菇粗多糖的表征及其抗氧化活性的比较

Characterization and antioxidant activity analysis on the crude Hericium erinaceus polysaccharides extracted by different methods

  • 摘要: 为探究不同提取方法对猴头菇粗多糖提取效果及抗氧化活性的影响。以猴头菇子实体为原料,通过热水、超声、碱液提取法分别提取猴头菇多糖,得到猴头菇粗多糖提取物:W-He P、U-HeP和A-HeP;研究和比较了不同提取工艺对多糖得率、理化性质和抗氧化活性的影响。结果表明:粗多糖得率依次为A-HeP(4.66%)>U-HeP(3.92%)>W-HeP(3.29%)。红外光谱分析表明3种粗多糖均呈现出典型的吡喃型葡聚糖和β-型糖苷键吸收;紫外光谱分析表明3种粗多糖组分中均含有蛋白质,而β-消除反应表明3种粗多糖组分中均存在O-糖肽键,说明3种粗多糖中的蛋白质为含有糖链和蛋白质残留的糖蛋白。对3种粗多糖组分的还原力和DPPH·、·OH、ABTS+·清除效果分析表明3种粗多糖均具有一定的抗氧化能力,其中A-HeP对DPPH·、·OH和ABTS+·均有较好的清除效果,其清除率分别可达到72%(5 mg/mL)、75%(5 mg/mL)和85%(0.5 mg/mL)左右。由此可见,碱液提取效果较好,碱提猴头菇粗多糖A-HeP具有较好的清除自由基能力,可作为天然抗氧化剂开发利用。 

     

    Abstract: To investigate the effects of various extraction methods on the crude polysaccharides from Hericium erinaceus and their antioxidant activities,three different methods,including hot water extraction,ultrasonic extraction and alkali extraction,were applied to extract polysaccharides from the fruiting body of Hericium erinaceus and three crude polysaccharides( W-HeP,U-HeP and A-HeP,respectively) were obtained.The yield,physicochemical characteristics and antioxidant activity of the three crude polysaccharides were investigated and compared.The results showed that 4.66% of the yield was obtained for A-HeP,while only 3.92% for U-HeP and 3.29% for W-HeP.IR spectrum analysis indicated that the three polysaccharides had typical absorption for pyran glucan and β- glycosidic bond.UV spectrum and β- elimination reaction further demonstrated the existence of protein and O- glycopeptide bond in these three polysaccharides,implying the existence of glycoprotein in these crude polysaccarides.Based on the analysis of the reducing power,scavenging abilities of DPPH,hydroxyl and ABTS~+radicals,all of the three polysaccharides showed certain antioxidant activity,especially A-Hep,which exhibited about 72%( 5 mg / mL) of scavenging effect for DPPH·,75%( 5 mg / mL) for ·OH and 85%( 0.5 mg / mL) for ABTS~+·. These results indicated that the alkali extraction method possessed better extraction effect. Meanwhile,A-HeP had excellent scavenging radicals ability,showing the potential to be a type of natural antioxidant.
