响应面优化双酶法提取川陈皮素工艺

Optimization of double enzymatic nobiletin extraction by response surface methodology

  • 摘要: 以柑橘皮为原料,建立紫外分光光度测定方法,研究响应面优化双酶法提取川陈皮素的最佳工艺。利用紫外分光光度法测定柑橘皮中川陈皮素的含量,在单因素实验基础上,探究纤维素酶和果胶酶复配比例、复合酶添加量、水浴温度、酶解时间、乙醇体积分数、料液比对川陈皮素得率的影响,并设计响应面分析优化方法。结果表明影响川陈皮素得率的强弱顺序为:乙醇体积分数>水浴温度>酶解时间>料液比。响应面优化后最佳工艺条件为纤维素酶和果胶酶复配比例1.5∶1、复合酶添加量5%、乙醇体积分数70%、水浴温度61℃、酶解时间2 h、料液比1∶15,所得川陈皮素得率为265.1μg/g。与响应面预测值相比,相对误差为2.3%,说明优化后得到的提取参数准确可靠,是一种适合于柑橘皮中川陈皮素提取的快速、高效方法。 

     

    Abstract: Citrus skin as raw materials,establishs the method UV spectrophotometric to study the best technique that nobiletin extracted by response surface double enzymatic. By UV spectrophotometry content of citrus peel Nobiletin,and on the basis of one factor test,explore cellulase and pectinase complex ratio of enzyme dosage,Water bath temperature,reaction time,concentration of ethanol,liquid ratio on nobiletin yield,response surface analysis and design optimization methods. The results show that the impact nobiletin yield strength of the order:ethanol concentration > water bath temperature > enzymolysis time > the material- liquid ratio. After the response surface optimization optimal conditions for cellulase and pectinase complex ratio of 1.5 ∶ 1,the amount of enzyme added 5%,the ethanol concentration was 70%,Water bath temperature was 61 ℃,the enzymolysis temperature time was 2 h,the material- liquid ratio was 1 ∶ 15,resulting nobiletin yield 265.1 μg / g. Compared with the predicted value of response surface,the proof test gained the free oil yield was 2.3%,which indicates that the extraction parameters optimized to give accurate and reliable,was suitable for citrus peel extract nobiletin fast and efficient way.
