二维液相色谱/双检测器法同时测定婴幼儿配方奶粉、米粉中维生素A、D3和E的含量

Simultaneous determination of vitamin A,D_3 and E in infant formula milk powder and rice flour by two-dimensional liquid chromatography / double detector

  • 摘要: 建立了在线二维液相色谱同时快速测定婴幼儿配方乳品和婴幼儿米粉中维生素A、D3和E含量的方法。首先,选择超高效C18柱和聚合键合的C18键合相PHA柱分别作为一维和二维分离柱,采用反相色谱法在一维上以甲醇、水作为流动相,二维上以乙腈、异丙醇作为流动相。采用四元泵作为一维分析泵,完成维生素A、E的定量和维生素D3的净化;根据维生素D3在一维色谱柱上的保留时间,确定切割时间窗口并以在线SPE柱收集含有维生素D3的馏分,另一个二元泵将SPE柱中的维生素D3馏分带到二维色谱柱中,完成维生素D3的定量分析;整个过程由两套色谱泵带动的二维色谱系统中运行,过程自动化程度高;只采用一个六通阀切换,检测时间短,只需要10 min就可完全分离所有目标峰。在上述优化条件下测定了婴幼儿配方奶粉和婴幼儿配方米粉中3种维生素的含量。通过本方法检测得到维生素D3的加标回收率为90.1%~99.7%,并与国标方法测定结果进行比较分析,结果表明,维生素A、D3和E偏差均小与<5%。表明该方法简单、快速、准确。 

     

    Abstract: The aim was to establish a simultaneous determination method for infant formula milk powder and infant rice in vitamin A,D_3 and E using line two- dimensional liquid chromatography.At first,to choose ultra- efficient C_(18) and C_(18) bonded polymeric bonded phase column PHA used as one- dimensional and two- dimensional separation column separately. One dimension with methanol and water acted as the mobile phase by reverse- phase chromatography method,and the two- dimension with acetonitrile and isopropanol as the mobile phase. Using quaternary LC pump as a one- dimensional analysis,complete quantitative of vitamin A,E and purification of vitamin D_3. The other two binary pumps put vitamin D_3 fraction in SPE column to a two- dimensional chromatographic column and to complete quantitative analysis of vitamin D_3. Run the entire two- dimensional chromatographic system consists of two pumps driven chromatography with high degree of automation,only a six- way valve was switched,and the detection time was short,only 10 minutes to complete separation of all the peaks.3 kinds of vitamins of infant formula and infant formula rice were detected under these optimal conditions.The standard addition recovery of vitamin D_3 by this method was 90.1% to 99.7%.And the measured results were compared with the national standard method that suggest that vitamin A,D_3 and E deviations were small and < 5%.It was showed that the method was simple,fast and accurate.
