冰岛刺参胶原蛋白多肽对PC12细胞氧化损伤的保护作用

Protective effects of collagen polypeptides from Cucumaria frondosa on oxidative damage in PC12 cells

  • 摘要: 目的:研究不同分子量范围的冰岛刺参胶原蛋白多肽C1(6 ku<Mr<10 ku)和C2(Mr<6 ku)对氧化损伤的神经细胞的保护作用及机制。方法:采用H2O2诱导高分化PC12细胞建立氧化损伤神经细胞模型。预先采用C1和C2处理细胞,浓度分别为0、12.5、25、50、100μg/m L,MTT法测定PC12细胞的增殖活性。将细胞分成正常对照组、模型对照组(H2O2)和样品保护组(胶原蛋白多肽+H2O2),采用不同浓度的C1和C2预处理细胞后,加入H2O2损伤细胞,分别采用试剂盒检测细胞内活性氧(ROS)水平、丙二醛(MDA)含量、培养基中乳酸脱氢酶(LDH)释放量、细胞内抗氧化酶和Caspase-3的活性。结果:当剂量大于50μg/m L时,C1和C2对H2O2损伤的PC12的增殖活性均有显著的提高作用(p<0.01)。高剂量组细胞内ROS水平均显著降低(p<0.05)。中、高剂量组细胞的LDH释放量和MDA含量均显著降低(p<0.01)、抗氧化酶(SOD、GSH-Px和CAT)活力均显著升高(p<0.05)。各组细胞凋亡因子Caspase-3的活性显著下降(p<0.05)。结论:冰岛刺参胶原蛋白多肽可以显著保护H2O2诱导的神经细胞损伤,其中低分子量的多肽效果更佳,可能与其氨基酸组成有关。 

     

    Abstract: Objective: To investigate the protective effects of collagen polypeptides with different molecular weights from Cucumaria frondosa on neural cells damaged by oxidative stress.Methods: Well- differentiated PC12 cells had been cultured in vitro,to establish an oxidative damage model of neural cells by hydrogen peroxide( H2O2).PC12 cells were pretreated with different concentrations of C1 and C2,accordance with the final concentration 0,12.5,25,50,100 μg / m L. The viability of the cells was measured by MTT method. Cells were then divided into the control group,damage group( H2O2) and collagen polypeptides protection groups( collagen polypeptides + H2O2).The reactive oxygen species( ROS),the content of MDA,the outleakage of lactic dehydrogenase( LDH),the activities of antioxidase and Caspase- 3 in PC12 cells were measured by corresponding kits. Results: When the dose was higer than 50 μg / m L,C1 and C2could promote the growth of PC12 injured by H2O2observably( p < 0.01).The level of intracellular ROS of high dose group cells were decreased significantly( p < 0.05).The LDH release,the content of MDA of middle and high dose group cells were decreased significantly( p < 0.01).The activities of SOD,GSH- Px and CAT of middle and high dose group cells were stimulated remarkably( p < 0.05). The activity of apoptosis factor Caspase- 3 was decreased significantly( p < 0.05).Conclusion: Collagen polypeptides from Cucumaria frondosa could play a neuroprotective role in PC12 cells oxidative damaged by H2O2.The more significant effect of the collagen polypeptides with low molecular weight correlated to their amino acids composition in all probability.
