超滤对大鲵多肽抗氧化活性的影响

Effects of UF (ultrafiltration) on the antioxidant activity of Andrias davidianus peptides

  • 摘要: 为了获得具有高抗氧化性的大鲵多肽,以大鲵肌肉为原料,经复合酶(风味蛋白酶和中性蛋白酶)酶解后,水解度达到44.12%,制得多肽得率为90.28%的大鲵多肽酶解液,用截留分子量分别为20、5、2、0.3 ku的超滤膜将其分离成5个分子量段,研究不同分子量段大鲵多肽对DPPH·、·OH和O-2·清除能力的影响。结果表明:不同分子量段的大鲵多肽组分的抗氧化能力大小顺序为:0.32 ku组分>25 ku组分>小于0.3 ku组分>520 ku组分。分子量为0.32 ku的大鲵多肽抗氧化活性最高,其对DPPH·、·OH和O-2·的EC50分别为0.4、0.7、1.5 mg/m L。因此,采用截留分子量为2 ku和0.3 ku的超滤膜可以分离得到具有高抗氧化活性的大鲵多肽。 

     

    Abstract: In order to obtain Andrias davidianus peptides with high antioxidant activity,combined- enzyme( flavourzyme and neutral protease) is selected to hydrolyze Andrias davidianus muscle.The degree of hydrolysis was obtained,up to 44.12%.The extraction rate of Andrias davidianus peptides was about 90.28%.The hydrolysate was separated into 5 peptide parts by ultrafiltration membranes with 20,5,2,0.3 ku respectively,the DPPH·,·OH,and O-2·scavenging ability of the different parts were studied respectively.The results showed that the strength of different parts' antioxidant capacity was in the order: 0.3~2 ku peptides > 2~5 ku peptides > peptides below 0.3 ku> 5~20 ku peptides.0.3~2 ku peptides had the highest antioxidant capacity,of which the EC50 of DPPH·,·OH,and O-2·scavenging ability was 0.4,0.7,1.5 mg /m L respectively.So ultrafiltration membranes with 2 ku and 0.3 ku were applied in the Andrias davidianus peptides with high antioxidant activity.
