小麦胚芽抗氧化肽对高糖诱导的血管平滑肌细胞增殖的保护作用

Protective effects of wheat germ antioxidant peptide on proliferation of vascular smooth muscle cells exposed to high glucose

  • 摘要: 目的:研究小麦胚芽抗氧化肽对高糖诱导的血管平滑肌细胞(vascular smooth muscle cells,VSMCs)增殖的保护作用。方法:用高糖诱导的VSMCs模型,将培养的VSMCs随机分为三大组:正常组(5.5 mmol/L葡萄糖)、高糖组(25 mmol/L葡萄糖,G)和实验组(25 mmol/L葡萄糖+不同浓度的抗氧化肽)。采用噻唑蓝(methyl thiazolyl tetrazolium,MTT)快速比色法和流式细胞仪检测抗氧化肽对VSMCs增殖活性及其细胞周期分布影响;通过酶标法测定在不同处理条件下细胞内总抗氧化能力(total antioxidation capacity,T-AOC)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)的含量变化。结果:小麦胚芽抗氧化肽可有效抑制高糖诱导的VSMCs增殖(p<0.01),阻滞细胞周期G1/S期转换,显著提高了细胞中总抗氧化能力,GSH-Px和SOD酶水平(p<0.01),降低了细胞中MDA水平(p<0.01)。结论:小麦胚芽抗氧化肽对高糖诱导的VSMCs增殖具有明显的保护作用。 

     

    Abstract: Objective: To evaluate the protective effects of wheat germ antioxidant peptide( AOP) on proliferation of vascular smooth muscle cells( VSMCs) exposed to high glucose. Methods: VSMCs were randomly divided into 3groups,normal control group( 5.5 mmol / L glucose),model control group( 25 mmol / L glucose) and treatment groups( 25 mmol / L glucose in different concentration of AOP).MTT( 3-( 4,5- dimethylthiazol-2- yl)-2,5- diphenyl tetrazolium bromide) assays were used to measure VSMCs proliferation,cell cycle distribution was analyzed by flow cytometry. The activities of total antioxidation capacity( T- AOC),glutathione peroxidase( GSH- Px),superoxide dismutase( SOD) and malondialdehyde( MDA) were measured by enzyme- linked immunosorbent assay.Results: VSMCs proliferation that was enhanced by high glucose was effectively suppressed by wheat germ AOP( p < 0.01),and the cell cycle G1/ S phase transition was also blocked. In addition,antioxidative enzyme( T-AOC,GSH- Px and SOD) activities in VSMCs exposed to high glucose were significantly increased in the presence of AOP( p < 0.01),whereas the MDA level was decreased( p < 0.01).Conclusion: These results indicated that wheat germ AOP exhibited significant protective effects on proliferation of VSMCs exposed to high glucose.
