响应面法优化参麦醒脑方提取工艺及益智作用初步观察

Optimization of extraction technology of shenmaixingnaofang by box- behnken design and observation nootropic effect of mice

  • 摘要: 目的:优化参麦醒脑方提取工艺并进行益智作用观察。方法:以党参多糖和党参炔苷得率为指标,通过单因素实验设计,考察超声时间、超声温度和料液比3个因素对提取工艺的影响趋势,采用Box-behnken响应面法对关键因素的参数范围进一步的研究和探讨;采用Morris水迷宫进行参麦醒脑方益智作用初步观察。结果:党参多糖和党参炔苷的最佳提取工艺参数:超声时间32 min,超声温度52℃,液固比1∶27,此条件下参麦醒脑组方中党参炔苷提取得率和党参多糖成分保留率分别为(5.18±0.22)μg/g和2.07%±0.15%。Morris水迷宫实验结果表明,参麦醒脑方能改善D-半乳糖、NaNO2致小鼠记忆损伤。结论:采用响应面法能够优化党参多糖和党参炔苷的最佳提取工艺;参麦醒脑方保健品具有益智作用。 

     

    Abstract: Objective: To optimize extraction technology and to observe the nootropic effect of Shenmaixingnaofang.Methods: With the transfer rates of lobetyolin and codonopsis pilosula polysaccharide,as evaluation indexes,the effect of the three factors,such as ultrasonic time,extraction temperature and solid to liquid ratio was investigated by the single factor experiment. By analysis of variance,the key factors that could influence the process were determined.Then the range of parameters of key factors was further studied and explored by Box- Behnken response surface methodology. The changes of learning and memorizing ability were tested with Morris water maze.Results: The optimum preparation conditions of the lobetyolin and codonopsis pilosula polysaccharide were as follows: ultrasonic time was 32 min,extraction temperature was 52 ℃,solid to liquid ratio was 1∶ 27.Under these conditions,the transfer rates of lobetyolin and codonopsis pilosula polysaccharide were( 5.18 ± 0.22) μg / g and2.07% ± 0.15%. Shenmaixingnaofang could improve the damage of learning and memory by D- gal and Na NO2.Conclusion: The optimum preparation conditions of the lobetyolin and codonopsis pilosula polysaccharide had been optimized by the response surface method,which could help to enhance the stability of the process. All the treatment groups diagnosed with Shenmaixingnaofang could improve the abilities of learning and memory in mice models.
