南极假丝酵母Pseudozyma sp.JCC207的培养及其海藻糖提取工艺优化

Optimization of culture conditions and trehalose extractive technology of Pseudozyma sp.JCC207

  • 摘要: 本文研究了南极假丝酵母Pseudozyma sp.JCC207的培养条件,对其海藻糖的提取工艺进行优化。利用单因素实验研究了培养温度和初始p H对南极假丝酵母生长的影响,采用RSM分析法确定了三氯乙酸(TCA)法提取南极假丝酵母海藻糖的最优工艺。结果表明其最适培养温度为25℃,起始p H为5~9范围内酵母生长情况差别不大;最优提取条件为:三氯乙酸浓度0.6 mol/L、体积15.20 m L、提取时间29 min;每克南极假丝酵母湿菌体中海藻糖的提取量为17.72 mg;功能实验表明,与甘油(20%)和二甲基亚砜(10%)等传统保种试剂相比,0.5%海藻糖溶液在短期内(5 d)对酵母具有更好的保种能力。 

     

    Abstract: The culture conditions of Pseudozyma sp.JCC207 from Antarctic and the optimization of the extractive technology for its trehalose were studied in this paper. Effects of culture temperature and initial p H of culture medium on Pseudozyma sp.JCC207 growth were explored by single factor tests,and the optimization of extraction condition of yeast trehalose by rchloroacetic acid( TCA) was determined using response surface method( RSM). The results showed that the optimum culture temperature was 25 ℃,and initial p H value of 5 to 9 impacted little on yeast growth,and the optimal extraction conditions were obtained as follows: TCA concentration 0.6 mol / L,TCA volume 15.20 m L,and extraction time 29 min. The extraction amount was 17.72 mg from one gram wet cells of Pseudozyma sp. JCC207.Function test indicated that 0.5% trehalose solution had a better conservation capacity for yeast in 5 days compared with 20% glycerol and 10% dimethyl sulphoxide( DMSO).
