硒化大米蛋白抗氧化性活性研究

Preparation of selenide rice proteins and their antioxidant activity

  • 摘要: 采用化学修饰法将酶解法获得的大米蛋白制备成硒化大米蛋白(Se-RP)并鉴定其抗氧化活性。研究表明制备的多种Se-RP均具有一定的谷胱甘肽过氧化物酶(GPX)活力,其中Se-RP24的GPX活力最高,约为116 U/g。各种Se-RP均可发挥清除1,1-二苯基-2-三硝基苯肼自由基(DPPH·)、羟自由基(OH·)和超氧阴离子(O2-·)自由基的作用,并可有效维持心肌线粒体的正常功能,抑制抗坏血酸/二价铁离子(VC/Fe2+)诱导损伤线粒体的膨胀,降低线粒体中脂质过氧化产物丙二醛(MDA)的含量。Se-RP的获得为大量制备植物源性硒化蛋白奠定实验基础,也为开发新型蛋白类抗氧化剂提供了新思路。 

     

    Abstract: Selenide rice proteins( Se- RP) were prepared via rice proteins obtained by enzymatic hydrolysis were chemically modified,then their antioxidant activities were identified. The results showed that Se- RPs possessed certain antioxidant activity of glutathione peroxidase( GPX),wherein GPX activity of Se- RP24 was highest,approximately 116 U / g. Andvarious kinds of Se- RPs could play a scavenging role in free radical such as 1,1- diphenyl-2- three nitrophenylhydrazine( DPPH ·),hydroxyl radical( OH ·) and superoxide anion( O).Furthermore,Se- RP could effectively maintain the normal function of cardiac mitochondria,inhibit swelling of damaged mitochondria induced by ascorbic acid / divalent iron ions( V_C/ Fe~(2+)),and decrease content of the lipid oxidation product MDA in mitochondria.Se- RP sobtained successfully laid the experimental foundations for large scale preparation of plant- derived selenide protein,and also provided new insight for the development of new albuminoidal antioxidant drugs.
