人工培养冬虫夏草胞外多糖的分离纯化及组成分析

Isolation,purification and composition analysis of extracellular polysaccharide from cultured Cordyceps sinensis

  • 摘要: 将冬虫夏草菌丝体培养液以醇沉法提取多糖,通过DEAE-52纤维素层析柱和Sephadex G-100凝胶层析柱分级纯化后得到两种单一的中性多糖组分EPS-1w和EPS-2w。应用紫外分光光度、Sephadex G-100凝胶柱层析法和比旋度法鉴定多糖的纯度,凝胶过滤法、理化实验和高效液相色谱研究其相对分子质量、理化性质及组成。结果表明:EPS-1w和EPS-2w为均一组分且易溶于水、稀酸、碱,其平均分子量分别约为4.65×104u和3.48×104u,均主要是由D-甘露糖、D-葡萄糖、D-半乳糖组成,其物质的量比分别为23.7∶6.2∶8.5和35∶3.6∶3.1。 

     

    Abstract: In the research,the polysaccharide from Cordyceps sinensis' mycelium culture had been extracted by alcohol sinking method.After been fractionated and purified with DEAE-52 cellulose column chromatography and Sephadex G-100 gel column chromatograph,two neutral polysaccharides were obtained which named EPS- 1w and EPS- 2w. UV- spectrophotometry,Sephadex G- 100 gel column chromatograph method and specific optical rotation method were used to identify the purity of these two polysaccharides. And the gel filtration method and physical and chemical experiments as well as high performance liquid chromatography had been used for analyzing the polysaccharides' relative molecular mass,physical and chemical characters and compositions. The results showed that both of EPS-1w and EPS-2w were homogeneous components,and they could be dissolved in the water,dilute acid and alkali.Their average relative molecular mass was about 4.65 × 104 u and 3.48 × 104 u.The EPS-1w and EPS- 2w were composed by D- mannose,D- glucose,D- galactose,and the amount of substance were 23.7∶ 6.2∶ 8.5 and 35: 3.6: 31.
