MALDI-TOF质谱法分析与鉴定骆驼乳中的磷酸肽

Analysis and identification of the phosphopeptides in camel milk by MALDI-TOF MS

  • 摘要: 建立一种基于基质辅助激光解吸电离-飞行时间质谱(MALDI-TOF MS)为基础的方法检测并鉴定骆驼乳中的磷酸化肽段。通过商品化的二氧化钛(Ti O2)材料富集驼乳中低丰度的磷酸肽,并优化出Ti O2富集材料与驼乳中磷酸肽相互作用最适宜的吸附洗脱条件,孵育缓冲液:乙腈/水(50∶50,v/v)+1%三氟乙酸,洗脱液为10%氨水。再将得到的磷酸肽溶液用MALDI-TOF MS方法检测,用Mascot数据库进行数据检索,能够从驼乳中鉴定出四个磷酸化肽段。本研究为不同哺乳动物乳中磷酸化肽段的检测提供了一些新的思路且能够应用于掺假乳的鉴别。 

     

    Abstract: The MALDI-TOF MS-based method was establised for determining and identifing phosphopeptides in camel milk. The low abundance phosphopeptides of camel milk was enriched using commercial Ti O2,and optimized the suitable adsorption-elution condition between the commercial Ti O2 and camel milk,incubation buffer:acetonitrile/water(50/50,v/v) +1% trifluoroacetic acid,eluent:aqueous ammonia(10%). The obtained phosphopeptides solution was determined by MALDI-TOF MS method,and the data searching was carried out using Mascot database,four phosphopeptides was identified in the camel milk. This study provided a new idea for the phosphopeptides of different mammal milk determination and applied to the adulterated milk identification in the practical application.
