大孔树脂纯化新疆圆柏总黄酮工艺研究

Purification of total flavonoids from juniperus sabina l.by macroporous resin

  • 摘要: 通过比较D101、AB-8、HPD400、HPD500、HPD417、HPD826六种大孔树脂的静态吸附效果,从中筛选出适合分离新疆圆柏总黄酮的树脂,并在单因素实验基础上正交优化最佳大孔树脂对新疆圆柏总黄酮的纯化工艺。结果表明,D101大孔树脂对新疆圆柏总黄酮具有较好的分离效果;最佳纯化工艺条件为,上样浓度1.2256 mg/m L,上样流速1.0 m L/min,除杂用水量5 BV,乙醇浓度50%,洗脱剂用量4 BV,洗脱流速1.0 m L/min。在此条件下获得总黄酮回收率为88.36%,纯度为69.96%。 

     

    Abstract: By comparing the static adsorption effect of D101,AB- 8,HPD400,HPD500,HPD417 and HPD826 macroporous resin,resin was screened out for separating total flavonoids from Juniperus sabina L.And purification process of total flavonoids from Juniperus sabina L. was optimized by orthogonal exceperiments based on single factor exceperiments. the results showed that D101 macroporous resin for total flavonoids had good separation effect.The optimal parameters for purification were as follows: the sample concentration of 1.2256 mg / m L,the sample flow rate of 1.0 m L / min,water consumption on removing impurity of 5 BV,ethanol concentration of 50%,eluting agent volume of 4 BV,desorption rate of 1.0 m L / min.Under these conditions total flavonoids had a recovery rate of 88.36% and a purity of 69.96%.
