黄绿蜜环菌菌种分离及人工培养研究

Study on the separation and domestication of Armillarialuteo-rivens strain

  • 摘要: 采用组织分离法从黄绿蜜环菌子实体的菌盖与菌柄结合处分离菌种,经多次纯化,获得纯化的黄绿蜜环菌菌株。以分离的黄绿蜜环菌母种为材料,通过单因素实验和正交实验,以菌落直径及菌丝体干重为指标,研究不同碳源、氮源、无机盐、维生素对菌丝体生长的影响。结果表明:黄绿蜜环菌菌丝最优化培养条件为马铃薯浸出液20%、黄豆芽浸提液10%、KH2PO40.3‰、VB1+VB21 mg/L(等比例),在此培养条件下,黄绿蜜环菌菌丝体干重可达6.5 mg/m L。 

     

    Abstract: With tissue-isolate method,the Armillaria luteo-rivens strain was isolated from the juncture between stipe and pileus of its fruit body. The purified strain was obtained after repeated purification. The strain as experimental material,the colony diameter and mycelia yield were taken as indexes to study the influence of mycelia growth by different carbon source,nitrogen source,inorganic salt,vitamin,and were carried out through the single factor test and orthogonal test. The result showed that the optimal culture condition of Armillarialuteorivens was potato 20%,extract of soybean sprouts 10%,KH2PO40.3‰,VB1+VB21 mg/L(proportion). Under this culture condition,the mycelia yield was up to 6.5 mg/m L.
