石榴花多糖醇提工艺的响应曲面法优化及其抗氧化活性研究

Study on optimization of extraction conditions for polysaccharide from pomegranate flowers and its antioxidant activities

  • 摘要: 以石榴花为原料,用热水浸提法提取其多糖。在单因素实验的基础上,运用Box-Behnken实验设计,采用响应曲面法对石榴花多糖的醇提条件进行优化。结果表明最佳工艺为:提取时间4h,温度76℃,料液比1∶52,在此基础上进行验证实验,实测多糖得率为6.35%。同时,不同的体外抗氧化实验结果表明石榴花多糖具有适度的还原力,较强的清除亚硝酸盐(NO2-)和超氧自由基(O2-)的清除效果,较好的螯合金属离子能力,而且石榴花多糖的抗氧化活性随着多糖浓度的提高而增强。 

     

    Abstract: Pomegranate flowers as row material were used for extraction polysaccharide with hot water. On the basis of single factor experiments,the optimized extraction conditions for polysaccharides were determined by using response surface methodology through the Box-Behnken design. The optional conditions were as follows:extraction time 4 hours,temperature 76℃,ratio of raw material to solvent 1∶52. Under the conditions,the yield of6.35% was obtained by the verification experiment. And the polysaccharides from pomegranate flowers showed moderate reducing power,strong clearance rate of nitrite(NO2-) and super oxygen free radical(O2-)and the ability to chelate metal ions,and the antioxidant activity of polysaccharides increased with the improvement of the polysaccharide concentration.
