黄连抗氧化活性组分的研究

Study on antioxidant activity of constituents from Coptis chinensis

  • 摘要: 为了解黄连抗氧化活性的物质基础,用水提醇沉法提取多糖,余液浓缩至干得浸膏,浸膏用高速逆流色谱法(HSCCC)按分配系数差异分离得总碱和亲水性组分,其中多糖含量用苯酚法测定,多酚含量用福林酚法测定;用清除线粒体ROS、DPPH·、O2-·及·OH法对各组分的抗氧化活性进行评价。结果表明,不同组分对ROS的抑制作用不同,均表现一定的抗氧化能力,线粒体ROS和DPPH实验说明浸膏抗氧化能力最强,IC50分别为1.37、0.62mg/mL,总碱对O2-·的清除作用最强,浸膏对·OH的抑制作用显著。以上结果表明黄连抗氧化作用是多组分共同作用的结果。 

     

    Abstract: In order to study antioxidant activity of constituents from Coptis chinensis extracts, High Speed Counter Current Chromatography ( HSCCC) was applied to separate extracts from Coptis chinensis according to distribution coefficient. The content of polysaccharide was determined determined with phenol-sulfuric acid method, and the content of total polyphenol was determined with Foline-Phenol method. Antioxidant activity of constituents was further evaluated by the methods of eliminating mitochondria-ROS, DPPH·, O2-· and ·OH-.The result indicated different constituents showed antioxidant activity, extracts showed strong oxidizing ability of eliminating mitochondria-ROS and DPPH·, IC501.37mg·mL-1and 0.62mg·mL-1, respectively. Total alkali showed obvious scavenging ability of O2-·. It seemed that antioxidant activity of Coptis chinensis were the result of coaction of all constituents.
