反相高效液相色谱法测定东方草莓中儿茶素的含量

Content determination of catechin in Fragaria orientalis Lozinsk. by RP-HPLC

  • 摘要: 建立东方草莓中(+)-儿茶素的含量测定方法。采用Agilent ZORBAX SB-C18色谱柱(4.6mm×150mm,5μm)分离;以乙腈-0.1%磷酸水为流动相进行梯度洗脱,流速为1mL/min,检测波长为280nm,对10批东方草莓中的(+)-儿茶素进行了含量测定。结果表明:(+)-儿茶素的保留时间为14.543min,其线性范围为0.05120.6400μg(R2=0.9999);加样回收率为99.70%(RSD 2.67%)。所建方法操作简单,稳定可靠,可用于东方草莓的质量评价。 

     

    Abstract: The purpose was to establish the method of ( +) - catechin in Fragaria orientalis Lozinsk. The samples were performed on Agilent ZORBAX SB- C18 ( 4.6mm × 150 mm, 5μm) column, eluted gradiently with mixture of acetonitrile-0.1% phosphate solution at a flow rate of 1mL /min, and detected at 280 nm.With this method, ( +) -catechin in 10 batches of F. orientalis Lozinsk. was determined. Results showed that ( +) - catechin retention time was 14.543 min.The calibration curves had good linearity within the range of 0.0512 ~ 0.6400μg ( R2= 0.9999) . The average recovery of ( +) - catechin was 99.70% ( RSD 2.67%) . The method was easy and stable, and could be used for the quantitative analysis of F.orientalis Lozinsk.
