乳酸片球菌PA003发酵培养基及发酵条件的优化

Optimization of fermentation medium and fermentation conditions for Pediococcus acidilactici PA003

  • 摘要: 为获得生产用乳酸片球菌廉价发酵培养基,采用单因素实验和正交实验的方法,对乳酸片球菌PA003的发酵培养基及发酵条件进行了优化。结果表明,最佳发酵培养基组成成分为:糖蜜2%、豆饼粉2%、玉米浸提液0.3%、磷酸氢二钾0.2%、乙酸钠0.5%、柠檬酸铵0.2%、硫酸镁0.058%、硫酸锰0.025%、吐温80 0.1%;最佳发酵条件为:接种量6%、pH6.5、温度37℃、培养时间14h、转速140r/min。采用上述培养基和培养条件进行培养,乳酸片球菌发酵液菌落总数为6.4×109CFU/mL,与采用MRS培养基培养时的菌落数相当,故显示出了良好的工业化应用前景。 

     

    Abstract: In order to obtain a low-cost fermentation medium for Pediococcus acidilactici PA003 in a large scale, the fermentation medium and fermentation conditions were optimized by single factor test and orthogonal test. The results showed that the best compositions of fermentation medium were 2% of molasses, 2% of soybean powder, 0.3% of corn extraction solution, 0.2% of dipotassium hydrogen phosphate, 0.5% of sodium acetate, 0.2% of ammonium citrate, 0.058% of magnesium sulfate, 0.025% of manganese sulfate, and 0.1% of Tween 80, and the pH of the medium was 6.5. The results also indicated that the best fermentation inoculum size was 6%, incubation temperature at 37℃ and time for 14h, speed at 140r/min. The biomass of Pediococcus acidilactici in such conditions reached 6.4×109CFU/mL, which was equivalent to the biomass in MRS medium.The results of this study suggested good application prospect in industry.
