高效液相色谱法检测蚕蛹油中主要脂肪酸的研究

Study on determination of main fatty acids in chrysalis oil by high performance liquid chromatography

  • 摘要: 建立了高效液相色谱-蒸发光散射检测器(HPLC-ELSD)同时检测蚕蛹油中三种主要脂肪酸的方法。以Hanbon Lichrospher C18柱(5μm,250mm×4.6mm)为固定相,丙酮-水(2%乙酸)(v∶v)为流动相,通过梯度洗脱实现目标产物的基线分离。针对各目标物建立工作曲线,亚麻酸、棕榈酸和油酸的线性范围分别为0.22.0mg/mL、0.12.0mg/mL和0.22.5mg/mL,R2>0.99,检出限(S/N=3)均为0.05mg/mL,回收率97%107%,相对标准偏差均小于2%。该方法快速、简便,可同时检测蚕蛹油中三种主要的脂肪酸。 

     

    Abstract: A rapid and simple method was established for simultaneous determination of three main fatty acids in chrysalis oil based on high performance liquid chromatography- evaporative light- scattering detection (HPLC-ELSD) . Chromatographic separation was carried out in gradient mode by using a Hanbon Lichrospher C18column (5μm, 250mm×4.6mm) with mobile phases of acetone-water (2% acetic acid) (v/v) . The fatty acids were in baseline separated, respectively. The calibration curves were plotted on the double logarithmic scales. The linear ranges of linolenic acid, palmitic acid and oleic acid were 0.2 2.0g/mL, 0.1 2.0mg/mL and 0.2 ~ 2.5mg/mL, respectively. R2>0.99, the limits of detection (S/N=3) were 0.05mg/mL, the recoveries were 97%~ 107% and the relative standard deviations were below 2%. The analytical method established in this work could be applied to accurate and rapid determination of three fatty acids in chrysalis oil.
