叠鞘石斛中联苄类化合物的体外抗氧化活性研究

In vitro antioxidant activity of bibenzyl compounds from Dendrobinm denneanum

  • 摘要: 将叠鞘石斛药材粉末经70%乙醇提取后,经HPD-300型大孔树脂纯化,得联苄类化合物。采用ORAC法、1,1-苯基苦基苯肼(DPPH)法、邻苯三酚自氧化法、fenton反应测定叠鞘石斛中联苄类化合物清除AAPH自由基、DPPH自由基、超氧阴离子自由基和羟自由基的能力。结果表明叠鞘石斛中联苄类化合物和V C清除AAPH自由基能力的ORAC值分别是0.33、0.21mg/L;叠鞘石斛中联苄类化合物和V C清除DPPH自由基的EC50分别是62.89、3.44mg/L;清除超氧阴离子自由基的EC50分别是408.10、14.04mg/L;抑制羟自由基的EC50分别是7.86、1.89mg/mL。与V C相比,叠鞘石斛中联苄类化合物具有较强的清除AAPH自由基的能力,能清除DPPH和超氧阴离子自由基,抑制羟自由基;其对自由基的清除能力在各用药浓度范围内呈一定量效关系。 

     

    Abstract: The 70% ethanol was used to extract the bibenzyl compounds from Dendrobinm denneanum, after HPD-300 macroporous resin purification, the bibenzyl compounds was obtained. The ability of removing AAPH·, DPPH·, superoxide anion radical and hydroxyl radical by the bibenzyl compounds solution were determined through oxygen radical absorbance capacity ( ORAC) , DPPH method, pyrogalbl autoxidation method and Fenton reaction.Results showed the activity of bibenzyl compounds from Dendrobinm denneanum and vitamin C with ORAC as index, the results were:0.33, 0.21mg /L ( scavenging AAPH·) , respectively.Antioxidant activity of bibenzyl compounds from Dendrobinm denneanum and vitamin C with EC 50 as index, the results were:62.89, 3.44mg /L ( scavenging DPPH) , respectively;408.10, 14.04mg /L ( scavenging O-2 ·) , respectively; 7.86, 1.89mg /mL ( inhibiting ·OH) , respectively.Compared with vitamin C, the clearance rates of AAPH·were higher in the bibenzyl compounds from Dendrobinm denneanum, could scavenge DPPH and superoxide anion radical and inhibit hydroxyl radical.The ability of removing radical showed a dose-effect relationship within the scope of the drug concentration.
