枯草芽孢杆菌K-6-9发酵条件优化及放大

Optimization of cultural conditions and amplification of Bacillus Subtilis-K-6-9

  • 摘要: 研究了枯草芽孢杆菌(Bacillus subtilis)K-6-9的发酵培养基、发酵条件。本实验通过单因素实验及正交实验得到的发酵培养基为:蔗糖1.0%,胰蛋白胨0.25%,牛肉膏0.5%,MgSO40.2%,KH2PO40.3%,CaCl20.1%。对摇瓶条件下K-6-9液体发酵的温度、转速、初始pH等主要影响因子进行实验探讨,确定了最佳培养条件为:初始pH7.0,装液量50mL/250mL,接种量2%,发酵温度37℃,转速180r/min,发酵时间18h。在摇瓶实验的基础上进行了5L发酵罐通气放大培养,K-6-9菌株14h达到生长高峰期,生长周期18h,此时所获得的芽孢量为1.40×109CFU/mL。相对于初始发酵培养基的芽孢量5.5×105CFU/mL提高了2.55×103倍。 

     

    Abstract: The single factor experiments and orthogonal experiments were adopted to optimize the liquid fermentation medium and conditions of Bacillus subtilis K- 6- 9. The results indicated that the constituents components of medium, the filling volume of shaking flask, the rotation speed and the temperature were the major factors which affected the biomass yield of K- 6- 9. The sucrose 1.0%, peptone 0.25%, beef paste 0.5%, MgSO 4 0.2%, CaCl 2 0.1% and KH 2 PO 4 0.3% were selected to be the medium formula, and the fermentation conditions were the filling volume 50mL in 250mL shaking flask, inoculum 2%, 37℃, shaking at 180r /min for 18h, and initial pH at 7.0.In the shake flask test was carried out on the basis of 5L fermentor enlarge cultivation, 14h reached the growth peak, growth cycle 18h, the optimal put pot for 14h, at this time of spore was up to 1.40 × 109CFU/mL, which was 2.55 × 103times higher than that of initial fermentation condition
