基于常压室温等离子体技术诱变选育D-乳酸高发酵速率菌株

Mutation and selection of high fermentation rate of D-lactic acid producing strain by atmospheric and room temperature plasmas

  • 摘要: 为了提高D-乳酸的发酵产率,以选育获得的D-乳酸高产菌Sporolactobacillus sp.Y2-8为出发菌株,采用常压室温等离子体技术对其进行诱变。利用高糖-溴甲酚绿高通量筛选平板及摇瓶发酵复筛,最终筛选获得一株具有良好遗传稳定性的D-乳酸高发酵速率突变株Y90s-1,其D-乳酸发酵产率达1.05g(/L·h),比出发菌株提高了36.3%。 

     

    Abstract: To obtain Sporolactobacillus mutants with improved fermentation rate, an atmospheric and room temperature plasma (ARTP) jet was firstly used to treat the Sporolactobacillus sp.Y2-8 which was bred in our previous research.The mutant with high D-lactic acid producing rate, named Y90s-1, was quickly screened according to high glucose concentration and pH indicator (bromocresol green) selecting plates.This mutant had good genetic stability and the D-lactic acid production rate reached 1.05g/ (L·h) , which was higher 36.3% than that of the original strain.
