超声和水提法提取灵芝多糖的结构和抗氧化性的比较研究

Structure and antioxidant properties of Ganoderma lucidum polysaccharides extracted by ultrasonic wave and hot water

  • 摘要: 分别采用超声和水提法制备灵芝多糖,并对两种多糖的性质进行对比分析。研究表明,超声提取和热水提取的灵芝多糖得率分别为2.16%±0.03%和3.26%±0.03%。超声提取多糖的分子量显著低于水提法灵芝多糖,同时通过原子力显微镜(AFM)观察,超声灵芝多糖的结构较水提多糖松散。此外,对两种灵芝多糖进行抗氧化活性对比发现,在多糖浓度为0.5mg/mL时,超声灵芝多糖的DPPH自由基清除率达到79.90%±0.30%,为水提法的1.3倍;多糖浓度为0.9mg/mL时,还原力达到1.035±0.001,为水提法的1.5倍;氧自由基清除能力(ORAC)为(2275.07±115.73)μmol Trolox/g,为水提法的1.8倍。 

     

    Abstract: Characteristics of Ganoderma lucidum polysaccharides extracted by ultrasonic wave and hot water were compared in this study.The yield of Ganoderma lucidum polysaccharides extracted by ultrasonic wave and hot water were 2.16% ±0.03% and 3.26% ±0.03%, respectively.However, the molecular weights of Ganoderma lucidum polysaccharides extracted by ultrasonic wave were smaller than those of polysaccharides prepared by hot water.AFM images showed that Ganoderma lucidum polysaccharides extracted by ultrasonic wave were looser.Moreover, antioxidant activities of two different polysaccharides were compared.The result showed that compared with the one extracted by hot water, the one extracted by ultrasonic wave was 1.3 times higher in DPPH radical scavenging activities (79.90%±0.30% at 0.5mg/mL) , 1.5 times higher in the reducing power (1.035±0.001 at 0.9mg/mL) , and 1.8 times higher in ORAC value (2275.07±115.73μmol Trolox/g) .
