苏芡种壳棕色素的提取及红外光谱分析

Extraction of brown pigment from the shell of Suzhou gorgon euryale seed and it's FT-IR spectroscopy analysis

  • 摘要: 以苏芡种壳为原料,采用溶剂提取法提取其中的棕色素,研究了不同提取剂、料液比、温度、时间等因素对棕色素提取效果的影响,通过正交实验确定了最适提取工艺条件。对所得棕色素粗制品分别用酸沉法和醇沉法进行纯化,并用红外光谱仪对所得色素Ⅰ和色素Ⅱ的结构初步进行了分析。实验结果表明:以2%NaOH溶液为提取剂、料液比1∶30、提取温度60℃、提取时间3h对苏芡种壳中的棕色素具有较好提取效果。色素Ⅰ和色素Ⅱ的红外谱图在3390.30、1633.44、1456.02、1338.38cm-1等处均出现较强的吸收峰,但色素Ⅰ和色素Ⅱ在2925.53、2854.18、1716.36、1234.24、867.82、815.75、777.18cm-1处吸收存在差异。这表明棕色素Ⅰ和棕色素Ⅱ结构中均存在酚羟基和芳香环等结构,但棕色素Ⅰ中存在着酯羰基结构,且具有C-N结构;而棕色素Ⅱ不存在这些结构,其芳香环上具有棕色素Ⅰ没有的取代基。 

     

    Abstract: Brown pigment was extracted from the shell of Suzhou gorgon euryale seed by using the method of solvent extraction.The influences of different factors such as the different solvents, the ratio of material to liquid, temperature and time on the extraction effect of brown pigment were compared. The suitable conditions of extracting brown pigment from the shell of Suzhou gorgon euryale seed were determined through the orthogonal experiment.The purified brown pigment were obtained separately with the method of ethanol- precipitation and hydrochloric acid-precipitation and its structure was analyzed by using a fourier transform infrared spectroscopy ( FT-IR) .The results showed that the suitable conditions of extracting brown pigment were as follows: 2% NaOH solution as extraction agent, material- liquid ratio 1 ∶ 30, extraction temperature 60℃, extraction time 3h. IR spectrogram of pigment Ⅰ and pigment Ⅱ obtained with the method of ethanol- precipitation and acid-precipitation revealed that 3390.30, 1633.44, 1456.02, 1338.38cm- 1were both the main absorption, but significant difference at 2925.53, 2854.18, 1716.36, 1234.24, 867.82, 815.75, 777.18cm- 1, etc.These indicated that brown pigmentⅠand Ⅱ both had functional groups including phenolic hydroxyl groups and aromatic rings structure. Brown pigmentⅠincluded the ester carbonyl structure and C- N structure, but brown pigment II didn't exist in these structures and had substituents in the aromatic ring which brown pigmentⅠdidn't contain.
