磁性Fe3O4/CS微粒固定超氧化物歧化酶的研究

Study on the immobilization of superoxide dismutase with magnetic Fe3O4/CS microparticles

  • 摘要: 采用磁性纳米Fe3O4为磁源,壳聚糖(Chitosan,CS)为高分子聚合物,利用凝聚/沉淀法制备磁性壳聚糖复合微粒(Fe3O4/CS),并进行了IR、VSM表征和粒度测试。采用交联结合法,以磁性Fe3O4/CS微粒为载体固定了超氧化物歧化酶(SOD)。研究了加酶量、固定化时间、pH、交联剂用量等对SOD固定化的影响。用正交实验法确定了SOD固定化的最佳条件:加酶量100mg,固定化时间240min,pH6.5,w=25%戊二醛用量0.20mL。固定化SOD重复使用4次后的酶活性及回收率仍能达到376U/g、56.5%,重复使用性能良好。 

     

    Abstract: In this thesis, using Fe3O4 as magnetic source and chitosan as polymer, magnetic chitosan composite microparticles (Fe3O4 /CS) were prepared by coagulating precipitation.The magnetic Fe3O4 /CS microparticles were charectered by IR, VSM, and the size distribution of magnetic Fe3O4 /CS was tested.Using the magnetic Fe3O4/CS microparticles as carrier, superoxide dismutase (SOD) was immobilized by chemical cross-Linking.Effect of the amount of SOD, time, pH and the amount of glutaraldehyde on the activity of immobilized SOD were studied.The optimum immobilization conditions of SOD were also studied by orthogonal experiment.The optimum immobilization conditions were as follows:the amount of SOD was 100mg per 200mg magnetic chitosan microspheres, the time was 240min, the pH was 6.5 and the amount of glutaraldehyde was 0.20mL.After using four times, the activity and recovery rate of immobilized SOD were 376U/g and 56.5% respectively, which showed that the immobilized SOD had good reusability.
