真姬菇多糖分子结构特性及生物活性研究

Structure characteristics and bioactivities of polysaccharides from Hypsizigus marmorous

  • 摘要: 以沸水浸提真姬菇(Hypsizigus marmoreus)子实体中粗多糖,用离子交换柱层析分离纯化得真姬菇纯化多糖HMP-3。经高效分子排阻色谱(HPSEC)分析,证明HMP-3为单一组分。单糖分析结果表明,HMP-3由葡萄糖和半乳糖组成,含量分别为80.18和10.82mg/g。采用高效凝胶尺寸排阻色谱-十八角激光散色仪-示差检测仪联用(HPSEC-MALLS-RI)联用技术,测得HMP-3的平均分子量(Mw)为(7460±110)ku,分子回旋半径值(Rg)为(21.6±0.4)nm。经甲基化分析和GC-MS检测,确定HMP-3主链主要由(1,4)葡萄糖组成,也含有少量的(1,6)葡萄糖或者(1,6)半乳糖。HMP-3虽然对人胃腺癌细胞(AGS)、结肠腺癌细胞(DLD-1)、子宫颈癌细胞(HeLa)没有明显的体外抑制作用,但免疫活性实验表明,该多糖可以促进小鼠巨噬细胞Raw264.7的增殖和剂量依赖性地提高巨噬细胞中NO的生成量,具有免疫调节活性。 

     

    Abstract: HMP-3 was isolated from Hypsizigus marmoreus fruit body by boiling water and fractionated by ion -exchange chromatography. HMP-3 was proved to be homogeneous by high performance size exclusion chromatography (HPSEC) . Their monosaccharide levels were significantly different, including the glucose content (80. 18mg/g) and galactose content (10. 82mg/g) . The average molecular weight (Mw) of HMP-3 was (7460±110) ku and its radius of gyration (Rg) was (21. 6±0. 4) nm determined by HPSEC-MALLS-RI system. The backbone of HMP-3 was mainly glucose molecules connected with 1, 4-glycosidic linkages which contained considerable amounts of glucose and/or galactose connected through 1, 6-glycosidic linkages. Little cytotoxicity was apparent in cancer cell lines (AGS, DLD-1, HeLa) from the addition of these polysaccharides;however, they could enhance the proliferation of murine macrophage Raw264. 7 cells and significantly stimulated Raw264. 7 to release nitric oxide (NO) , suggesting their potential immunomodulating activities.
