“黑美人”土豆色素对D-半乳糖诱导衰老小鼠抗氧化能力的影响

Effect of anthocyanin from Black Beauty Potato on antioxidation abilities of D-galactose induced subacute aging mice

  • 摘要: 研究"黑美人"土豆色素(Black Beauty potato anthocyanin,BBPA)对D-半乳糖诱导的亚急性衰老小鼠氧化损伤的保护作用。将昆明小鼠(32±2)g随机分为对照组,衰老模型和低(50mg/kg·bw)、中(100mg/kg·bw)、高剂量(200mg/kg·bw)花色苷共5组,100mg/kg·bwD-半乳糖颈背皮下注射建立小鼠衰老模型,同时灌胃给予不同剂量的花色素苷,对照组和模型组分别灌胃等体积的生理盐水,每天一次,连续给药30d后,分别测定小鼠体质量,血清中SOD酶活力、总胆固醇(TC)和总甘油三脂(TG),肝、脑组织中T-AOC、SOD酶活力及MDA含量。结果表明,与对照相比,D-半乳糖能显著降低小鼠体重(p<0.05)、血清中SOD酶活、肝脏和脑中GSH-Px酶活性,而血清中TG显著升高(p<0.05)。与模型相比较,花色苷处理组显著升高了小鼠体重、血清中SOD酶活性、肝脏中GSH-Px酶活性(p<0.05),以及脑中T-AOC、SOD、GSH酶活性(p<0.05),同时降低了肝脏和脑中MDA含量(p<0.05),其它实验结果组间差异不显著(p>0.05)。提示"黑美人"土豆花色苷具有一定的抗氧化能力,且具有靶向性,对脑组织的抗衰老作用较强。 

     

    Abstract: To study the effect of the protection of anthocyanin from Black Beauty potato (BBPA) by D-galactose induced subacute aging mice’s oxidative injury.Kunming mice were randomly divided into normal control, model, BBPA low dose treated (50mg/kg.bw) , medium dose treated (100mg/kg kw) and high-dose treated (200mg/kg·kw) 5 groups.Senile model were established by D-galactose hypodermic in jection for 30 day, the different doses of BBPA had been administered to three therapy groups.Normal control and aging model groups were administered with saline water.After 30 day, the mice body weight, the activities of SOD, total (TC) and total (TG) in the serum;then the activities of T-AOC, SOD, GSH and MDA content in liver and brain were measured respectively.Compared with normal control group, body weight, activity of SOD in the serum, activity of GSH-Px in liver and brain obviously reduced and TG increased (p<0.05) by D-galactose induced aging mice.BBPA treated groups significantly increased body weigh, the activities of SOD in the serum and GSH-Px in liver and T-AOC、SOD、GSH-Px in brain (p<0.05) , and significantly decreased MDA contents in liver and brain (p<0.05) .This data suggested that BBPA had good antioxidant effect.The targeted effects on brain was much more significant than those on serum and liver.
