高效液相色谱法同时测定绿原酸及其五种主要代谢产物

Simultaneous analysis of chlorogenic acid and its five major metabolites compounds by high performance liquid chromatography

  • 摘要: 建立一种同时测定绿原酸、咖啡酸、3-羟基苯丙酸(3-hydroxyphenylpropionic acid,3-HPA)、p-香豆酸(p-coumaric acid,PA)、阿魏酸(ferulic acid,FA)和马尿酸(hippuric acid,HA)含量的高效液相色谱方法。色谱柱为美国Agilent EclipseXDB-C18(250mm×4.6mm,5μm);流动相为甲醇和1%的冰醋酸水溶液,梯度洗脱;检测波长为320nm和253nm;流速为1mL/min。六种酚酸物质在一定范围内有良好的线性关系,相关系数为0.9979~0.9999,平均回收率为86.84%~101.56%,相对标准偏差为1.15%~2.29%。结果表明该方法快速、准确、灵敏度高,可用于绿原酸及其代谢物的检测。 

     

    Abstract: A method had been developed for the simultaneous determination of the contents of chlorogenic acid, caffeic acid, 3-hydroxyphenylpropionic acid, p-coumaric acid, ferulic acid and hippuric acid by high performance liquid chromatography.Six phenolic compounds were separated on a Agilent Eclipse XDB-C 18 (250 ×4.6mm, 5μm) column using CH 3 OH and 1% acetic acid aqueous solution as mobile phase with near gradient elution, and detected by ultraviolet detector (UV) at wavelength of 320nm and 253nm.There were good linear correlations between the concentrations and peak areas of the six phenolic compounds with the correlation coefficients of 0.9979~0.9999.The average recoveries were 86.84%~101.56% with relative standard deviations of 1.15% ~2.29%.The results suggested that this method was characteristic of rapidity, accuracy, high sensitivity.The method can be used for qualitative detection of chlorogenic acid and its metabolites.
