红茂草生物碱提取工艺优选及色谱分析

  • 摘要: 目地:优选红茂草生物碱提取与干燥工艺,对提取物进行分析鉴定。方法:优选提取方法,利用L9(34)正交实验优选红茂草生物碱的提取影响因子,高效液相色谱层析分离鉴定主要成分,用真空冷冻干燥技术进行干燥保存。结果:提取方式优选超声波提取法,L9(34)正交实验得液料比30∶1、pH7、乙醇体积分数75%、温度80℃为最佳的提取条件,平均得率可达4.215%,平均回收率和RSD分别为99.57%、0.68%,稳定性良好。异紫堇碱与原阿片碱含量显著高于其他生物碱。结论:超声波提取、真空冷冻干燥工艺适合于红茂草生物碱的提取,异紫堇碱和原阿片碱的高效液相色谱鉴定可用于红茂草生物碱制剂质量控制的指标。 

     

    Abstract: Objective:To get the alkaloids’ optimization of process of Dicranostigma leptopodum, detach and identify the extracts and dry storage. Methods:First the methods of extracting were choosen, then impact factors were selected by the L9 (34) orthogonal experiment, which was combined with high performance liquid chromatography, and the vacuum freeze-drying technology was used to store. Results:The optimal extraction means was ultrasonic wave extraction, and L9 (34) orthogonal best extraction conditions was liquid ratio 30∶1, pH7, 75% ethanol volume fraction and temperature 80℃. The average yield was 4.215%. The average recovery and RSD were 99.57% and 0.68%, with good stability. The content of protopine and Isocorydine were higher than other alkaloids. Conclusion:The technology of ultrasonic wave extraction, vacuum freeze-drying and HPLC were applied to extract, dry and separating inspection. The result was suitable for its extraction and identification.
